| Objective To compare the pharmacodynamic components of Zuojin Pill formula granule and traditional decoctions by HPLC(berberine hydrochloride,evodiamine and rutacarpine).A rat model of chronic gastritis will be established to compare the efficacy of Zuojin Pill formula granule and traditional decoctions in the treatment of chronic gastritis in rats.To evaluate the pharmaceutical chemistry and pharmacodynamic equivalence of Zuojin Pills’ traditional decoction and formula granules.Methods Through the naked eye to observe Zuojin Pills formula granules solution and its decoction of the same batch,to compare the color of both;using a pH meter to determine the pH value of both;qualitative comparisons of Zuojin Pill formula granule and its decoction by TLC method have been made;The HPLC method was used to determine the active ingredient content of Zuojin Pills formula granules and its decoction.The contents of berberine hydrochloride,coptisine hydrochloride,evodiamine and rutacarpine were compared.The rat model of chronic gastritis was established using hunger-satisfaction,aspirin suspension stimulation,and Helicobacter pylori stimulation.The experiment was divided into 4 groups:normal control group,model group,Zuojin Pill formula granule group and its decoction piece group,10 in each group.The gastric mucosal damage of pathological sections were observed by HE staining.The levels of inflammatory factors,EGF,SOD and MDA in the serum of the rats were measured.The effects of Zuojin Pills formula granule and its decoction on chronic gastritis in rats were compared.Results This article first compared the color and pH value of Zuojin Pill formula granule with its decoction pieces and found that the former has a darker color and a slightly higher pH than the decoction pieces;and the HPLC method was used to perform precision,repeatability,and stability identification of the Zuojin Pill formula granule and its decoction pieces,and to compare the content of berberine hydrochloride,berberine hydrochloride,evodiamine and rutacarpine.The results showed that the content of the components contained in the formula granules was higher than that of the pieces.Secondly,rat gastritis model was successfully established through hunger-satisfaction,aspirin suspension stimulation,and Helicobacter pylori stimulation.Pathological sections were observed by HE staining to observe the gastric mucosal damage,showing that the normal group had complete and smooth structure,and the model group had mucosal shedding,congestion and edema,and varying degrees of inflammatory cell infiltration.After drug treatment,the phenomenon of mucosal shedding was alleviated,and the therapeutic effect of Zuojin Pill formula granule was slightly better than that of its decoction.The detection of IL2,IL-6,IL-1β,IL-10,EGF,SOD and MDA activity by chemical colorimetric assay showed that the contents of IL2,IL-6,IL-1β,EGF and MDA in gastric mucosa of rats significantly increased,IL-10,SOD vitality decreased obviously.After administration,the levels of IL2,IL-6,IL-1β,EGF and MDA decreased,and the activities of IL-10 and SOD increased significantly.There was no significant difference between Zuojin Pill formula granule group and decoction group(P<0.01).Conclusion Zuojin Pills formula granule and decoction both contain effective chemical constituents of Zuojin Pills,and the content of active ingredients in formula granule is higher than that of decoction.Zuojin Pill formula granule and its decoction pieces have similar efficacy in the treatment of chronic gastritis in rats. |
