| Objective:To observe the expression of Chicken ovalbumin upstream promoter-transcription factor Ⅱ(COUP-TFⅡ)and Neuropilin-2(NRP2)in human gastric carcinoma and analyse the relationship between their expression and clinicopathologic features.Besides,we detected the relationship between COUP-TFⅡ and NRP2.Methods:Paired tumorous and adjacent non-tumorous human gastric tissues were obtained from 60 patients with GC who underwent surgical resection.We detected the expression of COUP-TF Ⅱ and NRP2 by Western blot and immunohistochemistry.In addition,the relationship between the expression of COUP-TF Ⅱ and NRP2 and clinicopathologic features was analied.We detected the relationship between COUP-TF Ⅱ and NRP2 by GV230 plasmid and siRNA infected SGC7901 by Western blot and RT-PCR.Results:According to the result of immunohistochemistry and Western blotting,COUP-TF Ⅱ and NRP2 were highly expressed within human gastric tumor specimens.The expression of COUP-TF Ⅱ was positively related to depth of tumor invasion,differentiated grade and lympth node metastasis(P<0.05).However,there was no significantly correlated with gender,age and the volume of tumor(P>0.05).Moreover,COUP-TF Ⅱ and NRP2 had a positively correlation in human gastric cancer(r=0.263,p=0.043).There was a significantly higher or lower expression of NRP2 when increasing or silencing the expression of COUP-TF Ⅱ in both gene and protein levels.Conclusion:The expression of COUP-TF Ⅱ is up-regulated in gastric cancer and has important value in determining invasion and metastasis of gastric cancer.COUP-TF Ⅱ can enhance the lympth node metastasis by stimulating expression of neuropilin-2.As a result,we suggest COUP-TF Ⅱ may be a new target to cure gastric carcinoma. |
