| Objective: To explore the effect of acupuncture on the contents of 5-HT,Glu and GABA sleep transmitters in the hypothalamus of PCPA rats after treatment and the effect of Glu/GABA ratio,and its effect on animal behavior of rats;To investigate the effect of acupuncture on insomnia in hypothalamus level of 5-HT,Glu,GABA and other sleep mechanisms.Methods: A total of 46 male SD rats were randomly selected from 10 normal groups,the remaining 36 were made and screened,the patients were randomly divided into model group,acupuncture group(navel inner ring acupuncture point combined with insomnia acupoint prescription)group,western medicine group(diazepam)group,10 in each group,and normal group,with a total of four groups.The acupuncture group was given acupuncture treatment of internal umbilical ring points and insomnia points by zhuang medicine,Western medicine group was given diazepam tablets solution gavage treatment,No treatment was given in the normal group and the model group,only the same time of scratch stimulation was given.After 6days of intervention,the mental state and body weight of the rats were observed,and the horizontal score,vertical score,center retention score,total distance of exercise,and fecal particle number of the rats in each group were detected by open-field experiment,he contents of 5-HT and GABA in each group were determined by elisa,and Glu in each group was determined by glucose assay.Results: 1.Observation of morphological behavior: the open-field experiment results showed that,compared with the normal group,the level score in the open-field of the model group showed an upward trend(P < 0.01),the central stay score showed a downward trend(P < 0.01),the total distance of exercise increased(P < 0.01),and the number of feces particles also increased(P < 0.05),Compared with the model group,the level score,the center stay score,and the total distance of exercise in the acupuncture group all decreased(P < 0.05),and the level score and fecal particle number also decreased,but there was no statistical significance(P > 0.05).2.The experimental indicators show that acupuncture and western medicine both have the effect of treating insomnia;Compare two by two;A、Compared with the model group,the content of 5-HT in the hypothalamus of the model group was significantly lower than that of the normal group(P < 0.01),the content of Glu was significantly increased(P < 0.01),and the content of GABA was significantly decreased(P < 0.01);B、Compared with the model group,the content of 5-HT in the hypothalamus of the diazepam group was significantly increased(P < 0.01).Glu content was significantly reduced(P <0.01).Significantly increased GABA content(P < 0.01)significantly decreased Glu/GABA ratio in valium group(P < 0.01);C、Compared with the model group,the content of 5-HT in the hypothalamus of the acupuncture group was significantly increased(P < 0.01),the content of Glu was significantly decreased(P < 0.01),and the content of GABA was significantly increased(P < 0.01);D、Compared with diazepam group,the content of 5-HT in hypothalamus of acupuncture group increased significantly(P < 0.01),no significant difference in the content of Glu and GABA(P > 0.05),and no significant difference in the Glu/GABA ratio(P > 0.05).Conclusions: 1.Conclusion of morphological behavior: acupuncture at navel inner ring point combined with insomnia prescription and diazepam group can relieve the tension and reduce the excitement of insomnia model rats,and the experimental results support that acupuncture is better than western medicine treatment.2.Conclusion of experimental indicators:A、The contents of 5-HT in the hypothalamus of PCPA insomnia rats were increased by acupuncture of navel inner ring point combined with insomnia prescription and diazepam group;B、Acupuncture at inner umbilical ring point combined with insomnia can reduce Glu content in the hypothalamus of PCPA rats with insomnia,increase the content of GABA,and reduce the Glu/GABA ratio in the rat model with insomnia;C、he effect of acupuncture is better than that of tranquilization,and the experiment supports the mechanism of this acupuncture intervention on moderate regulation of central sleep transmitters. |
