| Objective: To observe the methylation level of thyroid histone in patients with autoimmune thyroid disease(AITD).Methods: Selection of hashimoto thyroiditis patients and normal healthy controls all the 4 cases,extraction of thyroid tissue,add specific enzymes,antibodies,respectively formed immune complex protein,DNA,and by sequencing chromatin precipitation method(ChIP-Seq)detecting thyroid tissues the expression level of histone methylation,comparison between case and control group differences in the levels of histone methylation.Result:(1)The methylation level of histones in thyroid tissues was analyzed and it was found that the methylation level of histones in the case group was lower than that in the control group.(2)The methylation of histones in the HT group was different from that in the normal control group,and the methylation level in the HT group was lower than that in the control group.(3)After functional enrichment analysis of the distribution of histone sites in the genome,it was found that methylated genes with differences in the expression of histone were mainly involved in the molecular function and were enriched in the ribosomal signaling pathway.Conclusion: By thyroid tissue in patients with autoimmune thyroid diseases of histone methylation of omics analysis to know: HT group compared with healthy controls of histone methylation differences,when analyzing the function of enrichment as you can see,mainly there are differences in the molecular function of methylation of the gene,they mainly on the ribosome signaling pathways.This study suggests that epigenetic mechanisms may be involved in the development of autoimmune thyroid diseases. |
