Objective:To study the improvement effect and mechanism of Yiqi Yangyin and Huatan Huoxue method on the establishment of silicosis rat model by tracheal infusion of silica(Si02)suspension.Methods:Ninety-six male SD rats were randomly divided into normal saline control group(group A),silicosis model group(group B),low-dose treatment group(group C),medium-dose treatment group(group D),and high-dose treatment.Group(group E),tetrandrine group(group F),each group 1-16 number.The model group,the low-dose group,the middle-dose group,the high-dose group,and the Hanjia group were each infused with a 0.5ml of Si02 suspension(concentration: 50mg/ml),and the control group was perfused with the same amount of normal saline.After 2 weeks of successful modeling,the normal group was given 5 ml of normal saline,and the low,medium and high dose groups were given different doses of Yifei Huaxian Decoction,once a day,and the same amount of tetrandrine in Hanjia group was administered by 50 mg/kg.After 3 days,8 rats died in 4 weeks and 8 weeks,respectively.HE staining and Masson staining were used to observe the changes of lung tissue in rats.The levels of TGF-β1,Smad3 and Smad7 in serum and lung tissues were detected by ELISA.Results:1.The pathological sections of the lung tissue of the model group were used to determine the successful replication of the silicosis rat model.2.Pathological changes of lung tissue: 1)Visual observation After 2 weeks of modeling,the lungs of the control group were soft and smooth,with a pale pink color and no nodular formation.The surface of the lungs of the rats was rough,the surface was swollen,the color was pale,the capsule was slightly tight,and there were scattered gray-white punctate nodules.After 4 weeks,the lungs of the model group and the treatment group were pale,pale and dark red.Slightly hard,visible scattered in the bleeding point,the surface is uneven,the surface of the lungs can be seen scattered in the grayish white nodules.After 8 weeks,the color of the lungs of the high-dose Chinese medicine group and the Hanjia group was dark red,and the pale area was reduced.The small piece of unevenness was observed,and the gray-white nodules on both lung surfaces were reduced compared with the model group.2)HE staining The lung tissue of the control group was intact,the alveolar structure was clear,the wall was thin,and the interstitial was basically free of inflammatory cell infiltration.There was no significant change after 4 weeks and 8 weeks.At 2 weeks,the rats in the model group showed a large number of inflammatory cell infiltration in the alveolar space and interstitial,and the alveolar structure was destroyed.The interstitial edema and widening showed small fibrosis and cell nodules.The degree of pulmonary fibrosis at 8 weeks The aggravation was obvious,and obvious formation of sacral nodules and fusion of nodules were observed.Compared with the model group,the degree of inflammatory cell infiltration in the alveolar cells of the treatment group was alleviated to a different extent,and the nodules in the lung tissue were smaller and smaller than the model group.3.Pulmonary index: Compared with the treatment group,the lung index of the rats in the high-dose group changed significantly,P<0.05,the difference was statistically significant.Compared with the model group,the lung index of the low-,medium-treatment and Hanjia groups did not change significantly.The lung index of rats in the high treatment group changed significantly,and the difference was statistically significant(P<0.05).4.Pulmonary fibrosis index: 1)Serum TGF-β1,Smad3,Smad7: 4 weeks and 8 weeks,compared with the normal group,the expression of serum TGF-β1,Smad3,Smad7 decreased in each treatment group and model group;compared with the model group,each treatment group TGF-β 1 The expression of Smad3 decreased and the expression of Smad7 increased.The difference was statistically significant(P<0.05).Compared with the Hanjia group,the expression of TGF-β1 and Smad3 decreased and the expression of Smad7 increased in the high-dose treatment group.Significance(P<0.05).2)TGF-β1,Smad3,and Smad7 in lung tissues: 4 weeks and 8 weeks.Compared with the normal group,the expression of TGF-β1,Smad3,and Smad7 in the lung tissue of each treatment group and model group decreased.Compared with the model group,TGF in each treatment group.The expression of β1 and Smad3 decreased,and the expression of Smad7 increased.The difference was statistically significant(P<0.05).Compared with the Hanjia group,the expression of TGF-β1 and Smad3 in the lung tissue of the high-dose treatment group decreased,and the expression of Smad7 increased.Statistically significant(P < 0.05).Conclusions:Yifei Huaxian Decoction can significantly improve the lung tissue structure and fibrosis degree of silicosis model rats,and delay the progression of silicosis.The mechanism may be that Yifei Huaxian Decoction inhibits TGF-β1 in TGF-β1/Smads signaling pathway and The expression of Smad3 up-regulates the expression of Smad7 protein and reduces collagen deposition,thereby delaying the development of silicosis.
