Objective :To study the expression and biological significance of long chain noncoding RNA FOXD2-AS1 in esophageal squamous cell carcinoma,and to explore the biological function and mechanism of FOXD2-AS1 in esophageal squamous cell carcinoma cells.Methods: First,we detected the expression of FOXD2-AS1,a long chain non-coding RNA,in 86 pairs of esophageal cancer tissues and corresponding normal esophageal tissues collected from Nanjing General Hospital of Chinese PLA by real-time fluorescence quantitative PCR.After obtaining the expression of FOXD2-AS1 gene in esophageal squamous cell carcinoma,we constructed the siRNA interference RNAs and shRNA of FOXD2-AS1,and then transfected siRNA into esophageal squamous cell carcinoma cell lines.At the same time transfected shRNA into experimental nude mice by transfection.The transfection efficiency was still detected by real-time fluorescent quantitative PCR.siRNA and shRNA with high transfection efficiency were transfected into cells and nude mice respectively,the expression of FOXD2-AS1 was down-regulated,and then The effects of FOXD2-AS1 on the biological characteristics of esophageal squamous carcinoma cells were tested by animal simulation experiments,thanswell invasion,MTT proliferation,cell clone,Edu,cell scratch,and westernblot test.Results: The expression of FOXD2-AS1 gene in esophageal squamous cell carcinoma was significantly higher than that in corresponding normal esophageal tissue.The area under the ROC curve of tumor tissue was 0.799,and the area under normal tissue curve was 0.371,(P < 0.05).After transfection and down-regulation of FOXD2-AS1 expression,the invasiveness ability,The migration ability and proliferation ability in the interference group were significantly lower than that of the empty vector control group.After transfection,the cells were injected into the mice and the expression of FOXD2-AS1 was down-regulated.The tumor growth in the interference group was significantly smaller than that in the empty vector control group,and the difference was significant(P < 0.05).Conclusion: FOXD2-AS1 may be a proto-oncogene of the esophageal squamous cell carcinoma.The increase in the expression of the tumor can promote the proliferation,invasion and migration of the tumors of esophagus.
