In chrysanthemum,there are kinds of active materials,which closely related to the needs of various living functions for maintaining life,such as antioxidant,liver,bacteriostatic,anti-inflammatory,analgesia and so on.However,like many other medicine food homology plants,it is lacking a deep and detailed study of these active ingredients.The researches of active substances from chrysanthemum are concentrated on flavonoids,saponins and volatile oils.And the study contents are limited to a sample rough extraction of similar substances and its corresponding bioactivity.There is a shortage of research in separation and purification,structural analysis and the activity of purified product.This paper is mainly study the Imperial Chrysanthemum ethanol extracts,with the exception of flavonoids and saponins etc.Making a systematic and comprehensive study in extraction,isolation and purification,physicochemical properties,molecular structure and antioxidant activity.I hope it could make positive effects in the development of imperial chrysanthemum.The main contents and results of the research are as follows:In this paper,I determinate the basic components of Imperial Chrysanthemum with GB and literature method,and the results show that the content of water was 6.14 ± 0.02%,the total soluble sugar was 23.96±0.22%,the protein was 14.88 ±0.35%,the fat was 8.76±0.28%,the reducing sugar was 3.40±0.05%,the ash was 8±0.15%,the crude fiber was 7.36 ± 0.05%.The extractum of Imperial Chrysanthemum was prepared by ethanol extraction and vacuum concentration.By using the integral of scanning curve as the evaluation index,the paper established a TLC scanning(TLCS)method for determination of target product content.Combine with single-factor and orthogonal experiments to make an optimization of extraction conditions about three target substances.And the results showed that the optimum extraction condition of CG-1:extraction time was 5h,ethanol concentration was 40%,ratio of material to liquid was 1:40,integral value of TLC was 23605.34 ± 1332.06;the optimum extraction condition of CG-2:extraction time was 7h,ethanol concentration was 40%,ratio of material to liquid was 1:40,the integral value of TLC was 25301.74±1516.25;and the optimum extraction condition of CG-3:extraction time was 3h,the ethanol concentration was 80%,the material liquid ratio was 1:40,and the TLC integral value was 30416.74 ±1439.22.CG-1&CG-2 and CG-3(chrysanthemum extract)were obtained by silica gel column chromatography.And the purity of CG-3 was detected by TLC and HPLC.CG-3 crystal was got from during the gradient cooling process of the hyperthermal supersaturated solution.The structure of CG-3 crystal is four arris and double cone type.The results of DSC and TGA show that the melting point of CG-3 was 150.22 C,and the thermal stability was good when the temperature below the melting point.The optical rotation of CG-3 was-39° measured by a polarimeter.The structure of CG-3 was analyzed by TLC,GC-MS,FT-IR,1H-NMR 13C-NMR,HMQC and ESI.The results showed that the structure of CG-3 contained a obvious infrared absorption peaks of sugar,the analysis of monosaccharide composition was glucose,and the configuration of glucoside bond was alpha.The molecule contains a six membered ring structure of sugar and a five membered ring structure.C and C are connected with ether bond in non-saccharide fraction.The relative molecular mass was 342.098,the unsaturation was 2,and the molecular was C12H22O9S.Finally,the antioxidant activity of CG-1&CG-2 and CG-3 was analyzed by in vitro antioxidant model.The antioxidant activity of CG-1&CG-2 was weak,and there is no concentration effect.CG-3 has the ability to scavenge ABTS free radical,DPPH radical,superoxide anion and hydroxyl radical,and the clearance of ABTS free radical and hydroxyl radical was similar to VC.But the other two kinds of free radical scavenging effect were slightly weaker than VC.All of them had concentration dependence.
