| objectives:To investigate the quantity and clinical significance of CD19+IL-10+B cell and its subsets in peripheral blood and tumor tissues of patients with colorectal cancer(CRC).Methods Enrolled 38 CRC patients underwent surgery as observation group,from Nov.2017 to Dec.2018.And peripheral blood,tumor tissue and para-cancer tissue of these patients were collected,fromwhich,proportionsofCD19+IL-10+Bcell,CD19+IL-10+CD24hiCD38hii B cell,CD19+IL-10+CD24intCD38int B cell and CD19+IL-10+CD24hiCD38-B cells in CD19+B cells were determined and detected by flow cytometry.37 healthy volunteers were set as control group,and the same cell types as observation group were detected with the same method in peripheral blood only.And Lymphocytes from the peripheral blood of the two groups were extracted.The expression level of IL-10 mRNA was determined by Real-Time fluorescence quantitative PCR.Data was represented by Mean±Standard deviation((?)±S)and data was processed by statistical software SPSS 23.0.T-test and chi-square test were used to analyze the data.P<0.05 was considered statistically significant,while P<0.01 was considered statistically significant.Results There was no significant difference in the average age,gender composition and other general data between the observation group and the control group(P>0.05).RT-PCR results showed that the expression level of IL-10 mRNA in peripheral blood lymphocytes of the observation group was higher than that of the control group,with statistically significant difference(P<0.05).Compared with early CRC(I+II),IL-10 mRNA content in middle and advanced stage of CRC(III+IV)was significantly increased,and the difference was statistically significant(P<0.05).The content of CD19+IL-10+B cells in peripheral blood and tumor tissues of the observation group was higher than that of control group,and the difference was obviously statistically significant(P<0.01).The content of CD19+IL-10+CD24hiCD38hi B cells in the peripheral blood of the case group was higher than that in the control group,and the difference was statistically significant(P<0.01).CD19+IL-10+CD24intCD38int B cells were slightly higher in the peripheral blood of case group than that of control group,and slightly higher in the tumor tissue than in the healthy tissue,but the difference was not statistically significant(P>0.05).Peripheral blood CD19+IL-10+CD24hiCD38-B cells in case group were lower than those in control group,and the difference was statistically significant(P<0.01).And the content of CD19+IL-10+CD24hiCD38-B cells in tumor tissues were slightly lower than that in control group,and the difference was not statistically significant(P>0.05).Compared with early-stage CRC,the contents of CD19+IL-10+B cells and CD19+IL-10+CD24hiCD38hii B cells in the peripheral blood were increased in middle and advanced stage of CRC and the differences were statistically significant(P<0.01).The expression of CD19+IL-10+B cells and subgroups had no significant relationship with gender,tumor location and CEA concentration(P>0.05).Conclusions The expression of IL-10mRNA was significantly increased in peripheral blood lymphocytes of patients with colorectal cancer,and was significantly higher in the middle and late stages than in the early stages,suggesting that IL-10 may be related to the occurrence and development of tumors.CD19+IL-10+B cells were significantly increased in the peripheral blood and tumor tissues of CRC patients and they are also higher in the middle-late-stage CRC.The main increasing subgroup of CD19+IL-10+B cell was CD19+IL-10+CD24hiCD38hi B cell.These results hint CD19+IL-10+B cell and CD19+IL-10+CD24hiCD38hi B cell may connect with tumorigenesis and tumor-progress of CRC. |
