Objective:To explore the mechanism of Chaoyi Shengyang Yiqi Decoction in the treatment of gastric ulcer,and to provide experimental basis for clinical practice,as well as Chaoyi effective prescriptions for the treatment of gastric ulcer.Method:1.The 60 rats were divided into six different groups randomly(normal control group,stress gastric ulcer model group,low-dose,medium-dose,high-dose treatment group of Shengyang Yiqi Decoction,and positive control group).2.The other rats were induced acute gastric ulcer model by gastric perfusion with absolute ethanol,except for the normal control group.The acute gastric ulcer model was induced by intragastric injection of absolute ethanol(0.5 mL/100 g each).3.Regular intragastric administration once a day for 14 consecutive days.High-dose group of Shengyang Yiqi Decoction:6g/kg,2ml of decoction.Medium-dose group of Shengyang Yiqi Decoction:3g/kg,2ml of decoction.Low-dose group of Shengyang Yiqi Decoction:1.5g/kg,2ml of decoction.The positive control group was given ranitidine hydrochloride capsules(reference daily dosage of adults weighing 60 kg,0.15 g/granule,twice a day).Dissolve the capsules in distilled water before administration,and the dosage was 40 mg/kg.d.The normal group was given 0.9%sodium chloride solution of equal volume.4.After fasting for 24 hours,the serum was taken,and the contents of serum transforming growth factor(TGF-?1),prostaglandin E2(PGE2),epidermal growth factor(EGF)and glutathione peroxidase(GSH-Px)were determined according to the reagent instructions.The ulcer index and ulcer inhibition rate were also determined.Result:1.Transforming growth factor:Compared with model group,the content of transforming growth factor in treatment group and positive control group was lower with statistical significance(P<0.01).The content of transforming growth factor in low-dose treatment group was higher than that in positive control group with statistical significance(P<0.01).2.Prostaglandin E2:Compared with model group,the content of PGE2 in treatment group and positive control group was lower with statistical significance(P<0.05).The content of prostaglandin E2 in the medium-dose and low-dose group was higher than that in the positive control group with statistical significance(P<0.05).3.Epidermal growth factor:Compared with model group,the content of EGF in treatment group and positive control group was higher with statistical significance(P<0.05).The content of epidermal growth factor in the high dose treatment group was better than that in the positive control group with statistical significance(P<0.05).4.Glutathione peroxidase:Compared with model group,the level of glutathione peroxidase in medium-dose and high-dose treatment group was higher with statistical significance(P<0.05).The glutathione peroxidase in the middle and high dose groups was slightly higher than that in the positive control group without statistical significance(P>0.05).5.Ulcer index and ulcer inhibition rate:The ulcer index of the medium-dose and high-dose Shengyang Yiqi Decoction group and the positive control group was lower than that of model group,with statistical significance(P<0.01).The ulcer index of medium-dose and high-dose Shengyang Yiqi Decoction group was slightly higher than that of positive control group,but there was no significant difference(P>0.05).Conclusion:Shengyang Yiqi Decoction has obvious protective effect on gastric mucosal injury in rats with gastric ulcer.Its mechanism may be related to the increase of prostaglandin E2,epidermal growth factor,glutathione peroxidase and the decrease of transforming growth factor in rats with gastric ulcer.
