ObjectiveTO study the effect of Fufei Gushenyin for chronic obstructive pulmonary disease(chronic obstructive pulmonary diseases,COPD)model rat bronchoalveolar lavage fluid(Bronchial alveolar lavage fluid,BALF)mucins5ac(Mucin5AC,MUC5AC)content,blood leukocyte count and classification count,lung tissue pathology morphology and the influence of the general situation of rats,To explore the effect of Fufei Gushenyin on the treatment of COPD in rats and its effect on the high secretion of airway mucus,provide experimental basis for the clinical application of Fufei Gushenyin,and provide new ideas for the development of treatment programs for COPD and traditional Chinese medicine.MethodsAfter adaptive feeding for one week,50 male Wistar rats were randomly selected as the normal group,and the remaining 40 weremade into COPD rat models.The COPD rat model was induced by smoking and adding lipopolysaccharide(LPS)airway perfusion.After modeling,2 model rats and 2 normal rats were randomly sacrificed and the whole lung tissue was taken for pathological sections to confirm the modeling success.The remaining rats were randomly divided into the model group and the high,medium and low dose groups of Fufei Gushenyin.Then,the rats in the normal group and the model group were given distilled water,and the rats in the high,medium and low dose groups were given traditional Chinese medicine of Fufei Gushenyin at the corresponding concentration.The general conditions of rats were observed and recorded during the period.Enzyme-linked Immunosorbent Assay(ELISA)was used to detect the content of MUC5 AC in the rat bronchoalveolar lavage fluid.The pathological and morphological changes of the rat lung tissue were observed under the microscope,and the total number of white blood cells and their classification were counted by traditional manual method.Relevant experimental data were statistically processed by SPSS23.0 statistical software.Results1.General information of rats: before modeling,all rats were healthy,with normal eating,drinking,urination,body weight,sensitive reaction,bright color,and no cough,sneezing,asthma orother respiratory symptoms;After the model building,the rats gradually developed respiratory symptoms in the early stage,such as poor appetite,constipation,yellowing,and decreased activity.In the middle stage,the rats were slightly adapted to it,eating and drinking water increased,and there was a phenomenon of irritability,irritability and group fighting.In the later stage,the rats’ respiratory symptoms increased,and they were obviously emaciated,listless and haggard.After the intervention of Fufei Gushenyin with traditional Chinese medicine,the general conditions of rats were improved to varying degrees,and the improvement was best in the medium dose group of Fufei Gushenyin,while there was no significant difference between the high dose group of Fufei Gushenyin and the low dose group of Fufei Gushenyin.2.Histopathological morphology of rat lung tissue: the normal group had normal appearance and size of lung tissue,normal and complete structures of lung tissue cilia,vessel walls and alveoli were observed under pathological section microscope,and no obvious inflammatory cell infiltration was observed in each layer of mucosa.Compared with the normal group,the lung tissue of the model group showed increased volume,expansion,greyish white color,poor elasticity,increased exudate in the bronchial cavity,and abnormal structural changes such as cilium loss,tube wallthickening,capillary hyperemia,pulmonary bullae and a large number of inflammatory cells infiltration were observed in the lung tissue under pathological section microscope.After the intervention of Fufei Gushenyin with traditional Chinese medicine,the rats in the model group showed different degrees of improvement in lung tissue appearance and pathological microscopic observation,and the improvement was most obvious in the medium dose group of Fufei Gushenyin,while there was no significant difference between the high dose group of Fufei Gushenyin and the low dose group of Fufei Gushenyin.3.Leukocyte count and classification count: compared with the normal group,the leukocyte count of the model group was significantly higher than that of the normal group(P < 0.01),and the neutrophil level was mainly increased.Compared with model group,Fufei Gushenyin high,medium and low dose group of rats blood leukocyte counts significantly lower than the model group(P <0.01),and mainly neutrophils decline,Fufei Gushenyin medium dose group had the greatest reduction in drinking(P < 0.01),the leukocyte count and classification countof the Fufei Gushenyin high dose group and Fufei Gushenyin low dose group of rats have no obvious difference(P > 0.05).4.The content of MUC5 AC in BALF: compared with normalgroup,model group rats of MUC5 AC content in BALF were significantly higher than normal group(P < 0.01),compared with model group,Fufei Gushenyin high,medium and low content of MUC5 AC in rat BALF dose group were significantly lower than the model group(P < 0.01),and Fufei Gushenyin medium dose group significantly(P < 0.01),Fufei Gushenyin high dose group and Fufei Gushenyin low dose group of rats have no obvious difference(P >0.05).Conclusions1.Fufei Gushenyin can resist airway mucus hypersecretion,and its mechanism may be to improve airway mucus hypersecretion by reducing airway mucin 5AC secretion.2.Fufei Gushenyin can significantly reduce the inflammatory reaction in COPD rats and repair lung tissue damage.3.Fufei Gushenyin can significantly improve COPD rats’ cough,expectoration,asthma and other respiratory symptoms,with the effect of resolving phlegm,removing phlegm and relieving cough.Its mechanism may be related to its anti-airway inflammation and anti-airway mucus hypersecretion.
