| Objective To study the therapeutic effect of Jianpi Huoxue Recipe on rats with NAFLD model;based on the changes of oxidative stress and lipid peroxidation related index CYP2E1,to explore the mechanism of Jianpi Huoxue Recipe on NAFLD,and to analyze the clinical treatment of Jianpi Huoxue Recipe The mechanism of action of alcoholic fatty liver.Methods1.Animal grouped There were 80 male Kunming rats.They were randomly divided into two groups:20 rats in the normal group and 60 rats in the model group.After successful replication at the end of the 8th week,the high-fat model group was randomly divided into 5 groups,the model control group,the positive drug control group,and the spleen-reinforcing blood group.There were 10 low,medium and high dose groups.2.Model preparation The rat model of NAFLD was induced by high-fat diet with 88% common feed,10% lard,and 2% cholesterol.After 8 weeks of model induction,10 rats in the normal group and the model group were sacrificed.The successful preparation of the model was observed.3.medical treatement After successful replication of the NAFLD rat model,drug intervention was started.Rats in the normal group and the model group were given 0.9% saline.The positive drug control group and the low,medium and high dose groups of Chinese medicine were given simvastatin.Chinese medicine is administered at different concentrations.The stomach begins on the 8th weekend and lasts for 4 weeks.4.Detection of various indicators After the rats were sacrificed,the following indexes were detected;serum TC,TG,AST and ALT were detected in each group;TC,TG,FFA,SOD and MDA contents in liver tissues were detected;the relative expression of CYP2E1 in rat liver was detected by real-time PCR.Western The relative expression level of CYP2E1 in rat liver was detected by blotting method.5.Pathomorphological section After tissue sectioning,HE and Sudan III staining were performed.The pathological changes of liver tissues of rats in each group were observed under microscope at 8 and 12 weeks.Result1.Changes of CYP2E1 in rat model of NAFLD model1.1 Lipid metabolism and liver function: After eight weeks of high-fat diet induction,compared with the normal group,the blood levels of TC,TG,AST and ALT were increased in the NAFLD model,and the liver TC and TG were increased in the NAFLD model.Statistically significant(P<0.05);1.2 Lipid peroxidation index: After 8 weeks of high-fat diet induction,compared with the normal group,the FFA content,CYP2E1 protein expression and m RNA expression in the liver tissue of the model group were significantly increased,the MDA content increased,and the SOD activity decreased.Statistically significant(P<0.05).1.3 HE staining of hepatocytes showed that after eight weeks of high-fat diet induction,compared with the normal group,the NAFLD model group showed a large number of small vacuoles in the liver cells,the cytoplasm color became lighter,the cells were arranged disorderly,and the hepatic cords were unclear.Sudan III staining showed that compared with the normal group,the hepatocyte structure of the NAFLD model group was disordered,the hepatic cords were not arranged neatly,and scattered orange-stained lipid droplets were observed in the cytoplasm,and the lipid droplets were irregularly distributed.2.Effect of Jianpi Huoxue Recipe on rat model of NAFLD2.1 Lipid metabolism and liver function: After four weeks of drug intervention,compared with the model control group,serum TG,TC,ALT,AST,liver TC,and TG were significantly lower in the positive drug control group and the high-dose group.The difference was statistically significant(P < 0.05).2.2 Lipid peroxidation index: After four weeks of drug intervention,compared with the model control group,the positive drug control group and the Chinese medicine intervention medium and high dose group had significantly lower FFA in the liver tissue;compared with the model control group,the drug intervention group liver tissue The content of CYP2E1 m RNA and protein,MDA were significantly decreased,SOD was significantly increased,and the difference was statistically significant(P<0.05).2.3 Hepatocyte HE staining showed that after four weeks of drug intervention,compared with the model control group,the lipid droplets were observed in the cytoplasm of the positive drug control group,and the vacuole area was significantly less than that of the model group.In the high and middle dose groups,the lipid droplets in the hepatocytes decreased,and the hepatic cell fat lesions were significantly reduced.The Sudan III staining showed that compared with the model control group,the orange stained lipid droplets in the positive control group were significantly reduced;In the middle dose group,the amount of orange-stained lipid droplets in the liver cells was significantly reduced,and the steatosis was significantly reduced.Conclusion1.The therapeutic effect of Jianpi Huoxue Prescription on NAFLD model rats is remarkable,and the therapeutic effect of high dose group of Chinese medicine is better than that of medium and low dose group.2.The mechanism of Jianpi Huoxue Decoction in the treatment of NAFLD rats may be related to the reduction of the expression of lipid peroxidation index FFA,CYP2E1,MDA and the increase of SOD in the liver of rats with NAFLD. |
