| Human induced plxuripotent stem cells(iPSc)are obtained with somatic cells reprogrammed by transferring and overexpressing four pluripotent transcription factors.They can multi-directional differentiate and self-renew,also can be induced from the patient’s somatic cells to reduce immunity.And there is no ethical issue at the source.Thus,iPSc is widely used in clinical research.The use of stem cells to establish disease models to explore the pathogenesis of disease is currently a hot spot,but its relationship with tumor cells has not yet reached a conclusion.In the treatise,we studied how human iPS cells effect proliferation,tumor formation and metastasis of human lung cancer A549 cells through nude mice tumor formation model.OBJECTIVE:To study the effects of human iPS cells on proliferation,tumor formation and metastasis of human A549 cells.METHODS:Nude mice model of subcutaneous tumor formation and tumor formation model of tail vein injection were constructed by injecting human lung cancer A549 cells and human iPS cells respectively.The GFP and RFP fluorescent reporter genes were used to observe cell metastasis,distribution and tumor formation by live fluorescence imaging.The distribution,morphology and structure of A549 cells and iPS cells in tumors and organs of nude mice were observed by experimental techniques such as cryosection and paraffin section.RESULTS:Tumor formation experiment of subcutaneous injection shows that iPS cells can make A549 cells show stronger tumorigenic ability,and tumor formation is faster and earlier;while iPS cells of the same concentration had no tumorigenic ability.At the same time,iPS cells can promote the proliferation of A549 cells on organs and cause organ damage.Tumor formation experiment 1 of tail vein injection shows that iPS cells were more likely to survive,proliferate and metastasize in nude mice in the presence of A549 cells.And,with influence of iPS cells,A549 cells are more likely to cause disease in the livers,lungs and kidneys of nude mice.Tumor formation experiment 2 of tail vein inj ection shows that iPS cells transfers to the kidneys and heart earlier than A549 cells and promoted the metastasis of A549 cells.On the five organs of heart,liver,spleen,lung and kidney,iPS cells promote the proliferation of A549 cells and make it more likely to form tumors with obvious boundaries from surrounding tissues.In the experiment,frozen sections of the organs are observed to show two kinds of fluorescence appear in the same region simultaneously,indicating a strong interaction between the two cells.Frozen sections of the tumor showed that the inside of the tumor is mainly composed of A549 cells,and iPS cells promoted and supported the outside of the tumor. |
