Objective:To investigate the changes and significance of platelet-derived growth factor(PDGF),transforming growth factor-β(TGF-β1),interleukin(IL-1),α-smooth muscle actin(α-SMA),superoxide dismutase(SOD),and malondialdehyde(MDA)in the process of radiation-induced lung injury in rats,in order to reveal the protective mechanism of compound Jiujie tea in the occurrence and development of radiation-induced lung injury.Methods: 120 Sprague-Dawley male rats were randomly divided into blank group,model group,low dose group,middle dose group and high dose group according to random number table.The rats in the blank group were only reared without other treatment;the other groups were given a single irradiation to the right lung.Serum and lung tissues were taken from the five groups of rats after anesthesia for 10 days,40 days,and 70 days after irradiation:(1)Gross observation of rat lung tissue was observed by gross observation method;(2)Enzyme-linked immunosorbent assay(ELISA)The levels of serum inflammatory factors TGF-β1,IL-1β and PDGF were detected by the method;(3)Hematoxylin-eosin(HE)staining and Masson’s trichrome staining(Masson staining)were used to observe the pathological changes of right lung tissue in rats.alveolitis and pulmonary fibrosis were scored;(4)immunohistochemistry detection of α-SMA expression,serum determination of MDA content andSOD activity.Experimental results:(1)Gross observation of lung tissue: The color of the lung tissue of the blank group was pink,soft and elastic,and the surface was smooth.Rats in the model group showed general changes in the 10 days after irradiation.There were bleeding spots on the surface of the lungs,and the lungs were slightly swollen.The experimental group was relatively light.The surface of the model group showed pust spots 40 days after irradiation,compared with the experimental group.obvious.Subsequently,70 days after the irradiation,the lungs were atrophied,the surface was uneven,the texture became hard,and the surface of the lungs showed gray-white cord-like and nodular fibrotic lesions.It is heavier than the experimental group.(2)HE and Masson staining showed that the alveolar wall of the lung tissue of the blank group was thin,showing a single layer,and the alveolar structure was intact and clear,with no pathological changes such as edema and inflammation.10 days after irradiation in the model group,there was no obvious thickening of the alveolar wall,mild expansion of the capillaries,small alveolar cavity,partial alveolar epithelial shedding,mild edema of the pulmonary interstitial,no obvious collagen fiber deposition;40 after irradiation Day,the alveolar wall and alveolar space gradually thickened,the alveolar cavity was smaller than before,the edema gradually increased,accompanied by visible inflammatory cell infiltration and a little collagen fiber deposition;70 days after irradiation,the alveolar wall and alveolar space were significantly thickened,localAlveolar disappeared,regional consolidation,partial alveolar cavity loss,alveolar septum showed obvious fibroblast proliferation,a large number of fibroblasts,and significant collagen deposition;(3)Pulmonary coefficient:There was no significant difference in the blank group between the time points in the group(P>0.05).The lung coefficient of the model group was higher than that of the blank group(P<0.05),and the low dose was IX.The tea group was lower than the model group(P>0.05),and the middle and high dose groups were lower than the model group(P<0.05).The lung coefficient of the model group was higher than the blank group(P<0.05)at 40 days and 70 days after irradiation,low,medium and high doses.The group was lower than the model group(P<0.05);(4)Enzyme-linked immunosorbent assay(ELISA)showed that the serum TGFβ-1 content increased compared with the blank group,and the TGFβ-1 content in the serum increased with time.In addition to the 40-day low-dose group,the content of TGFβ-1 in the other groups was lower than that in the model group,and the difference was statistically significant.Serum Il-1content,at each time point after irradiation,compared with the blank group,the content of the model group increased P<0.05,reached the highest value in 40 days,and then decreased compared with the model group.The content of Il-1in the middle-high dose group was lower than that in the model group(P<0.05).The content of high-dose group in Jiujie tea was lower than that in the model group(P<0.05).The high-dose group of Jiujie tea was lower than the model 70 days after irradiation.Group,and the difference was statistically significant.The content of PDGF in serum was higher than that in the blank group.The content of PDGF in the model group was lower than that in the model group(P<0.05).The low dose group was lower than the model group 10 days after irradiation(P<0.05).The difference was statistically significant.(5)Changes inSOD and MDA contents in rat lung tissue Compared with the normal group,the SOD activity in the lung tissue of the model group decreased and MDA increased.There were significant differences in SOD between the high-dose group and the high-dose group at 10 days and 40 days after irradiation(P>0.05).There was no significant difference in the SOD activity in the high-dose group at 70 days after irradiation(P>0.05).P<0.05),there was no statistical difference in the low-dose group(P>0.05).There was no statistical difference(P>0.05)in the low-dose group 10 days after irradiation,10 days,40 days,70 days in the high school after irradiation.The MDA content in the low dose group was statistically different(P<0.05).(6)Immunohistochemical α-SMA staining showed that normal lung tissue was negative or weakly positive after refraction,and there was no statistical difference between the groups.At 40 days after irradiation,the expression of α-SMA in the lung tissue of the model group increased,and the muscle formation Fibroblast proliferation,mainly distributed in the bronchial and perivascular,compared with the drug-treated group(P<0.05),the normal group was lower than the model group(P<0.05);70 days after irradiation,the expression of sma in the lung tissue increased significantly.A large number of positive cells were observed under light microscope,which were in the form of strips or irregularities,mainly distributed in the fibrotic lesions.The model group was positively expressed in the medium and strong,and the expression of sma was increased compared with the drug group(P<0.05).The expression was higher than the normal group(P<0.05).Conclusion: Compound Sarcandra glabra can reduce the content of serum inflammatory factors,improve the degree of inflammation and fibrosis in lung tissue,increase the body’s antioxidant capacity,and hope to become a protective agent for radiation-induced lung injury.
