| EFFECT OF DAPHNETIN ON BIOLOGICAL FUNCTION OF HUMAN EEDOMETRIAL CANCER CELLSObjective: To investigate the effects of daphnetin on the biological function of human endometrial cancer cells HEC-1A and HEC-1B.Methods: HEC-1A and HEC-1B cells were divided into different concentrations of daphnetin treatment groups(25、50、75、100、200μmol/L)and control group.After 24,48,72 hours of culture,the cell counting kit-8(CCK-8)method were used to detect the proliferation rate.Then HEC-1A and HEC-1B cells were divided into different concentrations of daphnetin treatment groups(50、75、100μmol/L)and control group,After 48 hours of culture,scratch test and Transwell assay were used to detect the migration and invasion ability of HEC-1A and HEC-1B cells,respectively.Results:(1)After treatment with HEC-1A and HEC-1B cells for 24,48,and 72 hours,the cell proliferation inhibition rate increased significantly with the increase of the concentration of the Daphnetin;After treatment with Daphnetin for 48 hours,the inhibition of cell proliferation is most obvious;(2)After treatment of HEC-1A and HEC-1B cells by Daphnetin,compared with the control group,the cell wound healing rate decreased significantly and decreased with the increase of Daphnetin concentration(P<0.001).(3)After Daphnetin treatment of HEC-1A and HEC-1B cells,compared with the control group.The number of transmembrane cells was significantly reduced with increasing concentration of Daphnetin(P < 0.001).Conclusions: Daphnetin could inhibit the proliferation,invasion and migration of human endometrial cancer HEC-1A and HEC-1B cells,and the effect was dose-dependent.EFFECT OF DAPHNETIN ON THE RELATIVE EXPRESSION OF MRNA AND PROTEIN OF TP53,KI-67,CK7 AND CERBB-2 IN HUMAN ENDOMETRIAL CANCER CELL HEC-1BObjective : To investigate the effects of Daphnetin on the molecular markers of human endometrial cancer cell HEC-1B,including the m RNA and protein levels of TP53,Ki-67,CK7 and Cerb B-2.Methods:HEC-1B cells were divided into control group and Daphnetin treatment group(50、100μmol/L).After 48 hours of culture,the m RNA and protein levels of TP53,Ki-67,CK7,Cerb B-2 were detected by real-time fluorescence quantitative PCR and Western Blot respectively.Results :(1)After treatment with Daphnetin,the relative expression of TP53 m RNA and protein in HEC-1B cells increased compared with the control group,and showed a certain increase with the increase of Daphnetin concentration(P<0.001);(2)After Daphnetin treatment Compared with the control group,the relative expression levels of m RNA and protein of Ki-67 and Cerb B-2 in HEC-1B cells decreased,and showed a certain downward trend with the increase of Daphnetin concentration(P<0.001).(3)After treatment with Daphnetin,the relative expression of CK7 m RNA in HEC-1B cells was increased compared with the control group(P<0.001),and the relative expression level of CK7 protein was not significantly changed(P >0.05).Conclusion : Daphnetin can promote the relative expression of TP53 m RNA and protein in HEC-1B cells,inhibit the relative expression of Ki-67,CK7 and Cerb B-2 m RNA,and inhibit the relative expression of Ki-67 and Cerb B-2 proteins,but after the treatment of Daphnetin,the relative expressionlevel of CK7 protein has no significant change. |
