In modern research,the various chemical constituents of Polygonum multiflorum have the functions of lowering blood fat,anti-atherosclerosis,anti-oxidation,anti-aging and anti-tumor.2,3,5,4’-tetrahydroxystilbene-2-O-β-D-glucoside in Polygonum multiflorum has anti-inflammatory and analgesic effects,possibly inhibiting local inflammatory mediators such as prostaglandins and serotonin Release,direct inhibition of PGE2 and COX-2 synthesis.Bone cancer pain is a typical cancer pain with the main clinical manifestations of Breakthrough,Onging Pain,Allodynia and Hyperalgesia.Clinically,the main analgesics are still opioids.The use of long-term opioids will not only cause a decrease in cell surface opioid receptors,but also serious adverse reactions such as respiratory depression,nausea and vomiting.Object In order to explore whether the active ingredient of Chinese herbal medicine Polygonum multiflorum had analgesic effect and other analgesic effects,a model of rats tibia bone cancer pain were established.The success of the models were verified from three aspects: behavioral changes,body weight changes and mechanical pain threshold changes.The rat model of bone cancer pain has been successfully established.The ginseng glycosides,emodin and catechins were administered to the rats.The mechanical pain threshold changes,behavioral changes and growth conditions were used to study whether the extract of Polygonum multiflorum had analgesic effects,how effective were analgesia.Methods1.Establishment and evaluation of a rat models of bone cancer pain Thirty-eight female Sprague-Dawley rats were randomly divided into three groups: group A was a normal blank control group,and group B rats were injected with 6 sterile saline.Group C rats were implanted with breast cancer cells(walker256)in the left hind leg.A total of 24 bone cancer pain models.The Paw Withdrawal Threshold(PWT)measurements were measured using von-Frey filaments and the up-and-down method were chosen to reduce the error.All rats were treated with left hind legs on the day before surgery,one day after surgery,the third day after surgery,the fifth day after surgery,the seventh day after surgery,the tenth day after surgery,and the fourteenth day after surgery.The pain threshold was measured and the behavioral changes of the rats were observed at the same time,the walking time of the foot,the number of toes of the operation side,and the changes of the body weight of the rats were observed to observe the eating condition and pay attention to the changes of hair color and the like.After the end of the measurement on the fourteenth day,one rat was randomly sacrificed,and the left hind leg bone tissue was stained with HE to observe the invasion and destruction of the tumor cells.2.Observing the effect of different ingredients of Polygonum multiflorum Thunb on mechanical pain threshold in rats During the feeding process,8 rats in group C showed death,so the last 16 rats remained.Sixteen female SD rats in group C were randomly divided into four groups of C1,C2,C3 and C4 according to the intragastric drugs,with 4 rats in each group.On the 14 th,15th and 16 th day after modeling,the rats in the C1~C4 group were given normal saline(dose of 2ml),stilbene glycoside(dose of 60mg/ml,2ml),emodin(dose of 60mg/ml,2ml)and catechin(dose of 60mg/ml,2 ml);the left foot pain threshold was measured before each gavage,0.5h after gavage,and 1.0 h after gavage.After the measurement,the behavioral changes were briefly observed,and the number of toes,the appetite and the color of the hair on the surgical side were observed.Results The results showed that from the 5th day of feeding the cancer pain models,compared with the groups A and B,the appetite loss and behavioral changes of the rats in the group C included the lameness of some rats,and the rats in the sputum bite operation increased.The rats became aggressive and even some rats died and the mechanical pain threshold decreased(P<0.05).On the 10 th day,the appetite of the rats in group C were significantly reduced,the hair became dark,the body weight remained unchanged or even reduced a little,the left hind leg The tumor were clearly visible and there were a significant obstacle to walking.On the 14 th day,the pathological tissue sections showed obvious destruction of bone,severe infiltration of cancer cells,and disordered structure of trabecular bone.The rats in groups A and B had normal appetite,no significant change in behavior,and no significant difference in mechanical pain threshold.The bone structure were normal(P>0.05).In the late stage of bone cancer pain,rats will be accompanied by severe bone destruction.Therefore,the intervention time were selected from the 14 th to 16 th day after the establishment of the models.In the three-day gavage experiment,the mechanical pain threshold of rats in C2 stilbene glycoside were higher than that of C1 normal saline and C3 emodin,and that were increased in C4 emodin group significantly(P <0.05).The mechanical pain threshold of the group C2 were significantly increased after 0.5h and 1.0h after gavage(P<0.05),but the mechanical pain threshold was 0.5h after gavage and 1.0h after gavage.There were no statistical significance.After the end of the stomach,the appetite of the group C2 improved,but the body weight did not change significantly.Conclusion1.The models of bone cancer pain in rats in group C were established successfully.The mechanical pain threshold,behavioral,body weight,appetite and appearance were significantly different from those in groups A and B.2.Stilbene glycoside could effectively improve the pain threshold of rats with bone cancer pain,relieve pain,reduce hyperalgesia,and the effect were stronger than emodin,catechin and normal saline.In summary,this study suggests: Polygonum multiflorum could effectively reduce bone cancer pain in rats,and its effective monomer component were stilbene glycoside.
