| The requirements for the biodegradability of surgical sutures and the flexibility of fibers used in clinical surgery increase with the development of human civilization,medical technology.Polydioxanone(PPDO),a kind of biodegradable polymer materials,is used in the field of the third types of medical devices with excellent performance.Due to the biodegradability of PPDO,it is suitable to prepare surgical sutures,which can reduce the inconvenience caused by secondary suture removal and the risk of surgical infection.However,the relatively poor softness of PPDO fiber limits its wider application.In this research,the differences in compatibility of four plasticizers,including acetyl tributyl citrate(ATBC),epoxidized soybean oil(ESO),cyclohexane-1,2-dicarboxylic acid isodecyl ester(DINCH),ethyl oleate,on the PPDO membrane were invested.The effects of four plasticizers on biosafety and plasticity of PPDO membranes were also studied.In addition,the effect of ATBC on PPDO fiber properties and degradation performance in vitro was further evaluated.Methods and results: 1.Optimization of plasticizer for preparation of PPDO filmsATBC,ESO,DINCH,ethyl oleate and PPDO were mixed in a certain ratio,and PPDO film with the same size and thickness was prepared by melt hot pressing.The compatibility,biosafety,and plasticity were tested to select the plasticizer that had the best impact on the performance of the PPDO film.The results showed: ATBC had the best compatibility.The biosafety of DINCH,ATBC,ESO and ethyl oleate was in line with the national standard.ATBC has the best plasticizing effect.Therefore,ATBC is a more suitsble plasticizer for PPDO and studied in the next step.2.Study on the influence of ATBC on the properties of PPDO fibersThe ATBC with gradient ratio of 1%,2%,3%,4% and 5% of PPDO was mixed with PPDO to prepare PPDO fibers by spinning machine.The knot strength per unit area,Young’s modulus and biological safety of PPDO fibers were tested,and the optimum ratio of ATBC was selected to improve the properties of PPDO fibers.The results showed that 4% ATBC had achieved the best effect on the plasticity improvement of PPDO fiber,and the knotting strength per unit area was in line with the national standard.Therefore,4% ATBC is selected for the next in vitro degradation study.3.Study on the effect of ATBC on the degradation of PPDO fibers in vitroThe PPDO fibers prepared by adding 4% ATBC and without plasticizer were placed in the environment simulated with human body pH and temperature,and the following parts were evaluated:(1)degradation time and the effect of ATBC on the retention rate and elongation at break of PPDO fiber;(2)the influence on PPDO intrinsic viscosity and crystallinity;(3)the change of pH value of degradation solution.The results showed that:(1)with the prolongation of degradation time,the strength retention rate and elongation at break of PPDO fibers in the ATBC group and the control group decreased;in the first week of degradation,the intensity retention rate of the 4% ATBC group was higher than that of the corresponding control group(p<0.05);the intensity retention rate of the 4% ATBC group was lower than that of the corresponding control group at 2 to 6 weeks of degradation(p<0.05);during the 1~6 week of degradation,the elongation at break of the 4% ATBC group did not change significantly compared with the control group;(2)the intrinsic viscosity of the control group and the 4% ATBC group gradually decreased with the prolongation of degradation time,and the 4% ATBC group was significantly lower within 4 weeks of degradation than that of corresponding control group(p<0.05);(3)with the prolongation of degradation time,the crystallinity of the control group and the 4% ATBC group gradually increased,and the 4% ATBC group was significantly higher than that of the corresponding control group(p<0.05);(4)Compared with the corresponding control group,there was no significant change in the pH value of the degradation solution in the 4% ATBC group during the degradation process.Conclusion: 1.Plasticizer ATBC improves PPDO performance.2.In the degradation process of PPDO fiber,the addition of ATBC can improve the crystallinity of PPDO,the retention rate of PPDO fiber and the elongation at break.3.PPDO film and PPDO fiber prepared by using ATBC as plasticizer are biodegradable polymer materials with excellent performance,and have broad application prospects in the field of preparation of new scientific research and medical products. |
