Effect Of Shaoyao Decoction On Intestinal Microflora Diversity In Rats With Ulcerative Colitis Based On 16srDNA High-throughput Sequencing Technology

Objective: To establish a rat model of ulcerative colitis and to explore the effect of Shaoyao Decoction on the quantity and structure of intestinal flora in rats after administration of Shaoyao Decoction,Mesalazine and antibiotics by 16 srDNA high-throughput sequencing technology,and to explore the effect of Shaoyao Decoction on UC by regulating intestinal flora.Methods: 50 SD rats were randomly divided into blank control group,model group,mesalazine group,antibiotic group and Shaoyao Decoction group.Rat model of ulcerative colitis was duplicated by 100 mg/kg of TNBS solution plus 50% ethanol 0.25 ml enema.The rats in the mesalazine group were given mesalazine suspension(21 mg/mL-1)orally once a day,and the antibiotic group was given ammonia.Benzylpenicillin(1 g.L-1),vancomycin(5 mg.mL-1),neomycin(10 mg.mL-1)and metronidazole(10 mg.mL-1)were intragastrically administered twice a day.Shaoyao Decoction group was intragastrically administered with Shaoyao Decoction Concentrate(1 g.mL-1)once a day.Each dose of Shaoyao Decoction plus saline was 3 ml.The blank group and model group were intragastrically administered with the same amount of saline,once a day.All rats were sacrificed 7 days after intragastric administration.The feces of each rat were collected before treatment and 7 days after treatment.The 16 srDNA high throughput sequencing technique was used to sequence the faecal flora of rats.Result: Compared with the blank group,the number of OUTs decreased,alpha diversity decreased significantly,beta diversity showed differences.The relative expression levels of OUTs decreased in Bacteroides,Thickwallchia and Clostridium,and increased in Microbes verrucous,Cyanobacteria,Proteus,Bacillus,Camphor and Campylobacter.Compared with the model group,the number of OUTs increased,the alpha diversity and beta diversity were significantly different in Shaoyao Decoction group.The relative expression levels of OUTs in microorganisms,microorganisms,tracers,soft-walled bacteria,bacillus and Lactobacillus decreased,while the relative expression levels in thick-walled bacteria,proteus,clostridium,delta-proteus,epsilon-proteus,clostridium,Campylobacter and desulfurizing Vibrio increased.Compared with the model group,the number of OUTs increased significantly in the mesalazine group,while the alpha diversity and beta diversity were significantly different.The relative expression levels of OUTs in the microorganisms of warts,soft-walled bacteria,clostridium,bacillus,Lactobacillus and verruca decreased,while the relative expression levels of OUTs in the microorganisms of Bacteroides,Proteus,Cyanobacteria,4C0d-2,Proteus alpha,Proteus epsilon,Bacteroides,Campyloids,Lactobacillus,YS2 and RF32 were relatively expressed.The content increased.Compared with the model group,the number of OUTs decreased,the alpha diversity of single sample decreased,and the beta diversity showed a decreasing trend.The relative expression levels of OUTs in sclerenchyma,bacteroides,verrucous microorganisms,cyanobacteria,trachomonas,clostridium,verrucous microorganisms,proteus,campylobacter,Clostridium and verrucous microorganisms decreased,while the relative expression levels in proteus,bacillus,beta-proteus,Clostridium and verrucous microorganisms decreased.The relative expression levels of Proteus gamma,Bacillus burkholdii,Enterobacter and Lactobacillus increased significantly.Compared with the mesalazine group,the number of OUTs in Shaoyao Decoction group was not significantly different,the alpha diversity was consistent,the beta diversity was decreasing,and the composition and content of intestinal flora were very similar.There was no significant difference between Shaoyao Decoction group and the blank group,mainly composed of bacteroides,Clostridium and sclerenchyma.Shaoyao Decoction has obvious regulating effect on intestinal flora,which is manifested in the increase of probiotics and the decrease of pathogenic bacteria and conditional pathogenic bacteria.It shows that Shaoyao Decoction can regulate intestinal flora balance and improve intestinal flora imbalance.After gastric administration of antibiotics in UC rats,the intestinal flora imbalance is aggravated compared with the model group,and the species composition abundance is significantly reduced.Shaoyao Decoction group has beneficial bacteria.The expression level was not significantly different from that of mesalazine group,and the species diversity was similar to that of mesalazine group,which was consistent with the overall regulation direction of mesalazine group.Conclusion: The imbalance of intestinal flora may be the inducement of the development of UC.Shaoyao Decoction may play a role in treating UC by regulating the diversity of intestinal flora.
