Objective:The difference of metabolites in gastric cancer peritoneal lavage fluid samples was studied by liquid chromatography-mass spectrometry(LC-MC)to search for important potential biomarkers for the diagnosis of peritoneal metastasis of gastric cancer.Methods:62 patients with gastric cancer were selected in the first Hospital of Jilin University,during August 2018 to December 2018,their clinical data and peritoneal lavage fluid were collected.The positive rate of exfoliative cytology of peritoneal lavage fluid for gastric cancer was 14.52%(9/62).Using qRT-PCR technology to test CEA mRNA of the samples,the data were analyzed by Comparative Delta-Delta Ct method.According to the above data,62 samples of gastric cancer were put in groups as follows:(1)There were 24 patients with serosal invasion(A group),and 38 patients without serosal invasion(B group).(2)There were 13 patients with peritoneal carcinomatosis(C group),and 9 patients without peritoneal carcinomatosis(D group).(3)CEA positive group(E group),CEA negative group(F group).LC-MC was used to detect the metabolic group of peritoneal lavage fluid in 62 patients with gastric cancer.Total IonCurrent(TIC)and Principal Component Analysis(PCA)were compared between A and B,C and D,E and F.The differentiating metabolites were screened by T-test,discriminant analysis,cluster analysis,ROC curve and logistic regression analysis.Result:Eight different metabolites were screened out in group A and B:sulfite,TG(54:2),Glyceraldehyde 3-phosphate,alpha-Aminobutyric acid,alpha-CEHC,Dodecanol,Glutamylalanine and 3-Methylpropionic acid.The three different metabolites in group C and D were sulfite,CL(63:4),PE-NMe(40:5).Fourteen different metabolites in group E and F were sulfite,Glyceraldehyde 3-phosphate,TG(54:2),alpha-Aminobutyric acid,alpha-CEHC,Glutamylalanine,retinal,3-hydroxysterol,Tetradecenoic acid,MG(21:0/0:0/0:0),tridecanoic acid,Myristoylglycine,Octadecanoic acid and TG(53:4).Among them,sulfite was the differentiated metabolite screened by three groups.Sulfite,Glutamyla-lanine,TG(54:2),alphaCEHC,alpha-Aminobutyric acid and Glyceral-dehyde 3-phosphatewere the differentiated metabolites screened by A/B group and E/F group.Conclusion:1.There were significant differences in metabolites of peritoneal lavage fluid between patients with serosa invasion and patients without serosa invasion;there were significant differences in metabolites of peritoneal lavage fluid between patients with peritoneal metastasis andpatients without peritoneal metastasis;there were significant differences in metabolites of peritoneal lavage fluid between patients in CEA positive group and patients in CEA negative group.2.The concentrations of sulfite were differentially expressed in group A and group B,group C and group D,group E and group F.The concentrations of sulfite,TG(54:2),3-phosphoglyceraldehyde,glutamyl alanine,alpha-aminobutyric acid and alpha-CEHC were differentially expressed in group A and group B,group E and group F.Infenence : Sulfite 、 TG(54:2)、 Glyceraldehyde 3-phosphate 、Glutamylalanine 、 alpha-CEHC and alpha-Aminobutyric acid may be important potential biomarkers for the diagnosis of peritoneal metastasis of gastric cancer.
