| Objective To establish mouse model of Kawasaki disease by Lactobacillus casei cell wall extract(LCWE),and to detect the changes of matrix metalloproteinases(MMP-2、MMP-9)and assess the coronary artery lesion(CAL)by echocardiography and pathological detection,in order to explore the connection.Methods Extracted LCWE and diluted to 2.5 mg/mL,then it was used for immunization of genetically predisposed mice to establish animal model of coronary artery.Eighty BALB/C mice(4-6weeks old)were randomly divided into the experimental group and control group,40 mice in each group.Experimental group were injected with LCWE0.4ml(1mg),control group were injected with PBS.Observed and recorded the general condition and the weight of the mouse.Orbital blood samples were collected respectively at the 1st,3rd,7th and 28 th day after administration,which were used to measure the concentration of MMP-2 and MMP-9.The heart and liver samples were collected for pathological section to investigate the histopathological changes.Results The BW,HW and HW/BW in the experimental groups were obviously lower than those in the control groups at the 3rd and 7th day(P<0.05),and the changes in the pathological sections prove the animal model was successful.The concentration of MMP-2 、 MMP-9 has gradually increased in the earlier stage in the experimental groups,and the level decreased significantly as for 28 st day,while the concentration of MMP-2、MMP-9 got no significant change in the control group(P<0.05),and the trend of MMP-2 increased faster than MMP-9,and the difference from the control groups were bigger than difference of MMP-9.Conclusion The mouse could be infected with LCWE;MMP-2、MMP-9 is a fine index to reflex the CAL of KD,MMP-2 might be used for the early clinical diagnosis of KD patient. |
