Objective:Guided by traditional Mongolian medicine theory,this study used modern molecular biology technology to study the therapeutic effect of Mongolian medicine Sugemule-3Decoction(Sugmule-3 Decoction,SD-3)on isoproterenol-induced cardiac hypertrophy in rats,and to explore the mechanism of endoplasmic reticulum stress in the treatment of cardiac hypertrophy in rats by SD-3.Method:1.In vitro experiment: Rat H9c2 myocardial cells were selected for the experiment.ISO was used to establish the model of H9c2 myocardial cell injury and hypertrophy.MTT method was used to detect the toxicity and effectiveness of SD-3 aqueous extract.Western Blot,ELISA and fluorescence staining were used to detect protein expression in cells.2.In vivo experiment: Wistar male rats were selected for in vivo experiment.The model group and the treatment group were subcutaneously injected with ISO for 7 days to establish the rat model of myocardial hypertrophy.After successful modeling,SD-3 was used to treat the rats with myocardial hypertrophy by gavage.After 4 weeks of SD-3treatment,cardiac function and protein expression in heart tissue/serum were detected by echocardiography,HE staining,Western Blot and ELISA.Result:1.In vitro,SD-3 can effectively reduce cell mortality,alleviate cell hypertrophy,increase the expression of Calumenin protein,enhance the antioxidant capacity of model cells and effectively inhibit the related indicators of endoplasmic reticulum stress pathway.2.In vivo experiments,SD-3 can effectively improve the cardiac function of rats in model group,alleviate myocardial hypertrophy and tissue lysis and necrosis of rats in model group,increase the expression of Calumenin protein in myocardial tissue of rats in model group,enhance the antioxidant capacity of rats’ myocardium,and effectively inhibit the related indicators in endoplasmic reticulum stress pathway.Conclusion:In vivo and vitro experiments showed that SD-3 could effectively improve cardiomyocyte hypertrophy in rats.The mechanism may be that SD-3 could regulate oxidative stress and endoplasmic reticulum stress by up-regulating the expression ofCalumenin protein,thereby affecting the occurrence of apoptosis.
