BackgroundHepatocarcinoma is a common clinical digestive tract tumor.Abrus cantoniensis Hance is a traditional Chinese medicine for both medicine and food.It has the effects of clearing away heat and detoxifying,protecting liver and diuresis,benefiting stomach and strengthening spleen.It works well in the treatment of hepatobiliary diseases.Studies have found that Abrus cantoniensis Hance has the functions of protectinga liver,anti-bacterial and anti-viral,anti-inflammatory,immune regulation,lipid-lowering and scavenging hydroxyl free radicals.However,as far as the anti-cancer research on total saponins of Abrus cantoniensis Hance is still rare,although it has been reported that various components in Abrus cantoniensis Hance have a certain degree of inhibition on the growth of liver cancer,its mechanism of action remains unclear.Therefore,the mechanism of action of Abrus cantoniensis Hance against hepatocarcinomais worthy of further study.ObjectiveIn order to provide a theoretical basis for further clinical trials,treatment and new ways of utilization of Abrt.us cantoniensis Hance resources,in this study,Human Hepatocarcinoma HepG2 cells were used as research objects.CCK-8 method and flow cytometry were used to explore the effects of total saponins of Abrus cantoniensis Hance cell cycle and apoptosis of Human Hepatocarcinoma HepG2 cell.thus to clarify the anti-tumor mechanism of the drug outside the body.Methods(l)Eight concentrations of total saponins of Abrtus cantoniensis Hance were applicd to HLuman Hepatocarcinoma HepG2 cells for 24 hours and 48 hours.and the cell inhibition rate was detected by CCK-8 method,and the IC50 oftotal saponins of Abrus cantoinicensis Hance was calculatced.(2)Fiow cytometry to detee the effet of total saponins of Abrus cantoniensis Hance cell cyele of Human Hepatocarcinoma HepG2 cells.(3)Effeet of total saponins ot Abrus cantoniensis Hance apoptosis ot HumanHepatocarcinoma HepG2 cells by flow cytometry.Results(1)with the increase of the total saponin concentration and the prolongation of the action time.the cell viability decreased significantly.indicating that it has atime and coincentration dependent effect on the proliferation inhibition of Humain Hepatocarcinoma HepG2 cells.When the action time is 48 hours.only 0.5mg/mL can makE the cell inhibitioin rate at 51.25%.and the inhibition rate is higher than 50%;When the concentration rises to 2mg/mL,tHe inhibition rate is as high as 86.99%.After regression analysis.the halfi-inhibition rate(IC50)at 24 hours,and 48 hours was 1.1427 mg/mL and 0.504/mg/mL,respecti-vely(2)With the increase of the total saponin concentraion,it can be seen that the prolportion of synthesis phase is gradually increased compared with the coltrol group.and the proportion of Gap 2 phase was gradually decreased comparecd with the control group.while tlhe proportion of cells in Gap 1 phase was not obvious.It indicates that the total saponins of Abrus cantoniensis Hance have the effect of blocking the Human Hepatocarcinoma HepG2 cell cycle in synthesis phase.(3)When the total saponin concentration of Abrus cantoniensis Hance was less than 0.5 mg/mL,the effect of drug-induced apoptosis was not obvious.When the total saponin concentration of Abrus cantoniensis Hance was more than 0.5 mg/mL,the effect of drug-induced apoptosis was significantly enhanced and increases with increasing concentration;When the total saponin concentration of Abrus cantoniensis Hance reaches 2mg/mL,the apoptosis rate can reach 25%.Therefore,the Human Hepatocarcinoma HepG2 cell apoptosis rate of total saponins from Abrus cantoniensis Hance after 24 hours showed no significant difference in apoptosis rate in the low dose group and significantly increased in the high dose group.Conclusions(1)The total saponin extract of Abrus cantoniensis Hance can significantly inhibit the proliferation of Human Hepatocarcinoma HepG2 cells with time and concentration dependent effects.(2)The total saponin extract of Abrus cantoniensis Hance has the effect of blocking Human Hepatocarcinoma HepG2 cell cycle in synthesis phase.(3)The total saponin extract of Abrus cantoniensis Hance can induce apoptosis of Human Hepatocarcinoma HepG2 cells,but its induction of apoptosis is only effective at a concentration higher than 0.5 mg/mL,and it is concentration dependent.
