Objective: To investigate the relationship between VIP and visual developmental plasticity and the effect of monocular visual deprivation on the expression of VIP in the visual cortex. Methods: Thirty health tricolor guinea pigs aged 3-4 weeks were randomly divided into two groups: 15 in the normal control group and 15 in the monocular deformity deprivation group. The right eye was covered by eye mask suture. All guinea pigs were raised in the same natural light environment for 12 h and 12 h day and night cycle. Before intervention,1 week and 2 weeks after intervention, test the diopter,PVEP(the pattern visual evoked potential, graphical visual evoked potential),randomly executed after each check 5 guinea pigs at the same, using immunohistochemical technology to guinea pigs of semi-quantitative detection visual cortex VIP.Result: The biological parameter of guinea pigs were compared at different time points before and after occlusion, and the difference was statistically significant(P<0.05).Before occlusion, there was no statistically significant difference in biological parameter of eyeball and visual cortex between different groups(P<0.05).With the extension of covering time, the weight of the two groups of young guinea pigs gradually increased, and the hypermetropia diopter gradually decreased. The latent period of PVEP P100 wave in both eyes was gradually shortened and the amplitude was gradually increased in the normal group. There was no statisrical difference in the latent period and amplitude of P100 wave in both eyes(P<0.05).The latent period of P100 wave in the right eye(occluded eye)was prolonged and the amplitude was decreased. The incubation period of P100 wave in the light eye(unoccluded eye)was gradually shortened and the amplitude was gradually increased, and the difference of P100 wave in both eye was statistically significant(P<0.05).The difference of right eye P100 wave between normal control group and form deprivation group was statistically significant(P<0.05).There was no significant difference in left eye P100 wave between normal control group and form deprivation group(P>0.05).Among them, the right eye of the podophilia deprivation group was compared with the normal control group and the left eye of the patients before and 1 week and 2 week after the occlusion. There was no significant difference in VIP immunohistochemistry between the normal control group and the form deprivation group(P>0.05).There were significant difference in immunohistochemistry between the normal control group and the form deprivation group in VIP of the right visual cortex, and the difference were statistically significant(P<0.05).There were significant difference in VIP immunohistochemistry between the right and left visual cortex in the form deprivation group(P<0.05).Conclusion: Amblyopia can be formed by monocular occlusion in critical period of vision development.PVEP examination plays an important role in the diagnosis of amblyopia. The expression of VIP in the model of monocular form deprivation amblyopia is different, suggesting that VIP may be in the formation of visual development process.
