Objective: Bregenin has many biological activities and is widely used in the treatment of cardiovascular diseases.To study the protective effect of breviscapine in the cultured primary neonatal rat myocardiocytes with ischemia reperfusion injury.Method: The primary cardiac myocytes were isolated from the heart of SD suckling mice one or four days after birth.After culture,the primary cardiac myocytes were randomly divided into three experimental groups.Each group was given a compound pore,which was repeated four times:(1)Control group;(2)A/R group;(3)Bre+A/R group.According to the following indicators,the experimental groups were tested: the activity of LDH,GSH-Px,SOD and the content of MDA were analyzed by colorimetry;The cell viability was measured by MTS;The percentage of apoptosis were detected by annexin V-FITC/PI double staining.Results: a.Compared with the control group,the survival rate of cells in A/R group was significantly lower(p<0.01),LDH activity in culture medium was significantly higher(p<0.01),apoptotic number was significantly increased(p<0.01),the activities of antioxidant enzymes SOD and GSH-Px were significantly decreased(p<0.01),and the content of MDA was significantly increased(p<0.01),indicating that the myocardiocytes A/R injury model was successfully constructed;b.Compared with A/R group,the cell survival rate of Bre treatment group(20,40,80 ugM)increased in a dose-dependent manner(p<0.01),LDH activity in culture medium decreased gradually(p<0.01),apoptotic cell number decreased significantly(p<0.01),the activities of antioxidant enzymes SOD and GSH-Px increased significantly(p<0.01),and the content of MDA was decreased remarkably(p<0.01).Conclusion: Cardiomyocytes protecton of bregeni against anoxia/ reoxygenation injury mediated by inhibiting oxidative stress.
