| Objective:The invasiveness of pituitary growth hormone adenoma is the subject of research on the invasiveness of integrin-linked kinase(ILK)in other tumors and the research status of pituitary adenomas.There are three different levels at the molecular,cellular and organizational levels.To investigate the relationship between the expression of ILK and the invasiveness of pituitary growth hormone adenoma,and to further discuss the possible signaling pathways of ILK-mediated invasion of pituitary adenomas.Methods:In order to investigate the relationship between ILK expression and the invasiveness of growth hormone adenoma,this study was mainly verified by two aspects.First,through strict screening of clinical tumor samples,and through WB and immunohistochemistry and other detection methods to further verify the expression of important protein molecules directly related to invasion of MMP2,MMP9 and other two types of tumor tissue,verified in growth hormone adenoma Invasive and non-invasive adenomas have different invasive ability;a series of detection methods are used to compare and analyze the difference of ILK expression between invasive growth hormone adenoma and non-invasive growth hormone adenoma,and fully certify ILK expression and pituitary growth hormone gland The relationship between tumor invasion ability;on the other hand,primary culture of fresh clinical tumor samples and the purchase of existing growth hormone adenoma cell lines on the market,again from the cells by ILK inhibitors and lentiviral transfection down-regulation Further verify the relationship between ILK signaling pathway and tumor cell invasion and migration ability.Results:Firstly,in the two groups of tumor samples screened,the WB and immunohistochemical results of the tumor samples also indicated that MMP2 and MMP9 were highly expressed,and the invasive group between the two groups wassignificantly higher than the non-invasive group.Meanwhile,the experimental results It also showed that there was also a difference in ILK expression between the two groups,and the invasive group was higher than the non-invasive group,which was consistent with the results of MMP2 and MMP9.Subsequently,based on the results of the tissue level,we further verified the results at the cellular level.In the primary cells of pituitary tumors,cellular immunofluorescence suggests that ILK is expressed in tumor cells.Transwell invasion and migration experiments show that the invasion and migration of primary tumor cells in the invasive group is significantly stronger than that in the non-invasive group,and ILK inhibitors can Significantly inhibited the invasion and migration of tumor cells;at the same time,we performed CCK-8proliferation experiments on primary tumor cells,but found that the primary cell proliferation index of the invasive group was not significantly different from that of the non-invasive group,but at Ki-67(greater than In 3%)high tumors,the proliferation index was significantly higher than that of Ki-67(about 1%);at the same time,we further verified that In the primary cells of the invasive group,ILK inhibitors also significantly inhibited the expression of MMP2 and MMP9 proteins.Finally,we repeated the above experiments in the purchase of fresh growth hormone adenoma cell lines,and ILK inhibitors and lentiviral transfection down-regulated ILK.Expression can significantly inhibit the invasion and migration of tumor cells,and also regulate the expression of MMP2 and MMP9 proteins in cells.Conclusion:ILK is expressed in both growth hormone adenomas and ubiquitously expressed in various types of pituitary adenomas,and is involved in the invasiveness of growth hormone adenomas.It may be involved in the regulation of tumor invasion by regulating the expression of MMP2 and MMP9. |
