| Objective:Bone tissue is the same as human life,it needs to undergo stages of growth,development,aging and lesions,old bone removal,the process of new bone formation is the reconstruction of bone.In our bodies a certain amount of bone is absorbed every day,there is a considerable amount of bone mass synthesis,But because of our growing age the human body is in decline,the absorption of bone gradually accelerates to break the original balance,bone mass in the human body gradually reduces bone density,this is where osteoporosis occurs(Osteoporosis;OP).Many Chinese medical scientists have demonstrated the close relationship between visceral and osteoporosis in the early years,especially in the kidney.In clinic,the clinical effect of reducing the occurrence and development of this disease has been achieved by means of the treatment of kidney and other diseases.Beyond that,recent studies have also found parathyroid hormone(PTH)can promote the absorption of calcium ions(Ca2+)in the blood,The calcium in the blood is converted to calcium in the bones,to promote bone growth and enhance bone strength and achieve the effective prevention of osteoporosis.Therefore,The purpose of this study is to explore the two aspects related to osteoporosis,The correlation between the polymorphism of TCM and parathyroid receptor gene polymorphism in postmenopausal osteoporosis and the TCM syndrome of postmenopausal osteoporosis.in this way,the correlation between different syndromes of osteoporosis patients and the distribution of related genes in China and the TCM syndromes of osteoporosis were analyzed,early screening for osteoporosis can reduce the risk of osteoporosis,reducing their harm to people improves people’s living standards.Methods:This study collected 300 postmenopausal patients from September 2015 to May 2016 in the people’s hospital of Shanxi Province,The mean age is 67.4(plus or minus5)years,The average age of menopause was 49.16(plus or minus 3.63),with an average height of 1.56(plus or minus 0.3)meters.And they are all normal postmenopausal patients,not the unnatural menopause caused by diseases and other causes.The patients who came to the clinic were treated with double X-ray determination of bone density in the hospital,The average bone mineral content,L2~L4 bone mineral content,left hip joint and right hip joint were determined.According to the determination of peak value judgment in postmenopausal osteoporosis(postmenopausal osteoporosis,PMOP)patients,records of age,height,weight,lifestyle,etc.A group of experts from the hospital’s associate directorof TCM and above has established a group of experts,based on the criteria of TCM clinical diagnosis and treatment terms,the questionnaire was developed.All patients were surveyed with four diagnostic parameters,and the TCM syndrome differentiation of each postmenopausal osteoporosis patient was obtained,and each patient’s questionnaire should be completed by the same TCM expert.For patients after group completion,draw blood according to different syndromes,DNA Blood kit(OMEGA)kit was used to extract peripheral venous Blood from patients,the whole genome was extracted.Premier5 software design primer,the upstream primer is 5’-cattctgtgtactatagttg-3’,downstream primer is5’-GAGCTTTGAA TTA GCA-3’.Under the condition of 94 ℃ degeneration for 1minute,under the condition of 55 ℃ annealing for 1 minute,under the condition of 72 ℃for 1 minute,Finally,in 72 ℃ under the condition of continue 7 minutes.Repeat 35 cycles,after that,polymerase chain reaction(PCR)was selected for amplification,the PCR products were observed using 1.5% agarose gel electrophoresis,the distribution frequency of genotype and allele was analyzed under the gel image analysis system.Conclusion:Through the experiment:(1)Among the 300 samples,there were a large number of osteoporosis patients,with 211 cases in total,accounting for 70.3% of the total.In the four types of syndromes,there were a total of 93 cases,with a total of 93 cases,accounting for 31% of the total,accounting for 44.1% of the total patients;The fewest blood stasis was the lowest in 21 cases,accounting for 7% of the total population and 9.9%of the total patients;(2)PTH genotype: BB type(387,213bp),BB type(600bp),BB type(600bp,387 bp,213bp.The genotype frequency of B and B was not statistically significant(P>0.05),there was no statistically significant difference between B and B alleles in four groups(P>0.05);(3)In the normal control group and the experimental group,the genotype frequency of PTH was not statistically significant between the two groups(P >0.05). |
