| Objective: To investigate the effect of Chaihu Shugan Powder on endoplasmic reticulum stress-related molecular,glucose-regulated protein78(GRP78)and c-Jun N-terminal kinase(JNK),in gastric antrum of functional dyspepsia(FD)rats.Methods: Thirty-six SPF rats were divided into normal group,blank model group,Chaihu Shugan Powder low concentration group,middle concentration group,high concentration group and domperidone group according to the random number table method,with 6 rats in each group.Except for the normal group,the other groups of rats were fed with modified "Guo’s" tail stimulus every morning and evening to prepare the FD model.Then,on the same day of modeling,different groups were given the corresponding aqueous solution of the drug,and the stomach was administered at a dose of 1.5 ml/100 g.And the low,medium and high doses of Chaihu Shugan Powder were 0.16 g/ml,0.32 g/ml and 0.64 g/ml,and domperidone 0.30 mg/ml.The model group was given normal saline gavage,this operation lasts 4 weeks.The changes of body weight,diet anddrinking water in rats were observed and recorded during the establishment of model.Observe changes in life habits and emotions,and determine whether there are piles,irritability,and changes in hair roughness and gloss.After 4 weeks,gastric emptying rate was measured by semi-solid paste gavage method.Then all rats were anesthetized to take blood and stomach to observe the surface of gastric mucosa,whether there were flushing,ulcer or erosion.The levels of serum GRP78 and JNK proteins were detected by Elisa,the expressions of GRP78 and JNK in antral tissues were detected by Real-Time PCR,and the expressions of GRP78 and JNK proteins in antral tissues were detected by immunohistochemistry.Results:(1)Comparisons of general observation results of rats: during the modeling period,rats in the model group appeared piled up,decreased activity,sleepiness,reduced dietary and drinking water,weight loss,hair dryness and other phenomena;The above-mentioned manifestations existed in different concentration groups of Chaihu Shugan powder and in domperidone group,but the degree was slight.Compared with the model group,the weight gain of rats in the low,medium and high concentration groups of Chaihu Shugan Powder and domperidone groups was not significantly different(P>0.05);compared with the normal group,the weight gain of rats in the model group was significantly reduced,with statistical significance(P<0.05).(2)After antral tissue dissection,there was a small amount of semi-solid paste in the stomach of normal rats,and the surface of gastric mucosa was intact,smooth and light red.In the model group,there were more semisolid pastes in the stomach and the gastric mucosa showed flushing,varying degrees,no obvious ulcer and erosion.The retention of semisolid paste in the stomach of rats in different concentration groups of Chaihu Shugan Powder and domperidone group was less than that in modelgroup,but more than that in normal group,and the flushing degree of gastric mucosa was lighter than that in model group.(3)Comparisons of gastric emptying rates in rats of each group:Compared with the normal group,the gastric emptying rate of the model group was significantly lower(P<0.05).Compared with the model group,the gastric emptying rate of the low,medium and high concentration group of Chaihu Shugan Powder and the domperidone group increased(P < 0.05).(4)Comparison of Elisa results of rats in each group:Compared with the normal group,the expression of serum GRP78 and JNK protein in the model group was significantly higher(P < 0.05),and the expression of serum GRP78 and JNK protein in the low,medium and high concentration group of Chaihu Shugan Powder and the domperidone group was significantly lower(P<0.05).(5)Comparison of Real-Time PCR results of rats in each group: Compared with the normal group,the expression of GRP78 and JNK in antral tissue of model group was higher than that of normal group(P<0.05);compared with model group,the expression of GRP78 and JNK in each medication group was lower(P<0.05).(6)Comparisons of immunohistochemical results of rats in each group: Compared with the normal group,the levels of GRP78 and JNK protein in antral tissue of model rats were significantly increased(P < 0.05);compared with model group,the expressions of GRP78 and JNK protein in antral tissue of rats in low,medium and high concentration of Chaihu Shugan Powder group and domperidone group were significantly decreased(P<0.05).Conclusion: Chaihu Shugan Powder can effectively promote gastric motility.The mechanism of Chaihu Shugan Powder in treating functional dyspepsia may be related to reducing the expression of GRP78,JNK protein and mRNA related to endoplasmic reticulum stress. |
