| Objective:To evaluate the anti-inflammatory and antioxidative effects of andrographolide on the rat air pouch model of inflammationMethods:The experiment was divided into six groups,blank group(Control): 1% CMC-Na;model group(AP): 1% CMC-Na;control group(AP+I): 30 mg/kg indomethacin;low dose group(AP+L): 30 mg/kg andrographolide;middle dose group(AP+M): 50 mg/kg andrographolide;high dose group(AP+H): 70 mg/kg andrographolide,except for the control group given subcutaneous injection,The remaining groups were administered by intragastric administration.The pouches were induced by injecting sterile air into the intra-scapular region of the rats followed by LPS(20 μg/kg)administration.And the blank group rats were injected with sterile balanced salt solution.The air pouch tissue was taken for HE pathological section,and the exudate cells were centrifuged to count the total number of leukocytes;the back lavage fluid was taken for analysis of antioxidant indices(MDA,GSH,SOD,CAT).Rat plasma was taken to determine the contents of TNFα,IL-6,MDA,GSH,SOD and CAT.Results: Compared with the model group,the leukocyte counts were down-regulated in each drug-administered group(P<0.05).With the dose of andrographolide increased,the levels of TNFα and IL-6 in the balloon lavage fluid and plasma of the rats in each drug-administered group were significantly decreased(P<0.05).Compared with the model group,the levels of SOD and GSH in the lavage fluid and plasma of rats in the middle and high dose groups of andrographolide were significantly increased(P<0.05);the levels of CAT in the lavage fluid and plasma of rats in the low and middle dose groups were increased(P<0.05);while the levels of MDA in the lavage fluid and plasma of rats in the middle and high dose groups were significantly lower(P<0.05).It can be seen from the air pouch pathological results that the inflammatory cells of the high dose group of andrographolide are significantly less than the model group inflammatory cells,and it is also superior to that of the control group.It can be explained that andrographolide has a good anti-inflammatory effect.Conclusion: It seems that andrographolide is anti-inflammatory and antioxidative in nature,by reducing the number of leukocytes in each experimental group,down-regulating the levels of TNFα and IL-6,lowering MDA and CAT levels,increasing SOD and GSH levels,and the middle and high doses have better oxidation resistance than low doses. |
