| Objective:The present study aims to investigate the correlation between epithelial-mesenchymal transition(EMT)and preoperative chemoresistance in gastric cancer.Method:(1)sample collection:56 patients with preoperative chemotherapy and gastric cancer surgery were enrolled by inclusion and exclusion criteria.(2)Experimental grouping:According to the sensitivity of gastric cancer patients to chemotherapy,they were divided into sensitive group(clinical re-mission)and drug resistance group(clinical stability or progression).(3)Sample treatment:Immunohistochemical staining(IHC)technique was used to detect the expression of EMT-related markers E-cadherin and N-cadherin in gastric cancer tissues of experimental patients.(4)Data analysis:Statistical analysis of experimental data was performed by using IBM SPSS Statistics 25 version statistical software.The expression of EMT was compared between the resistant group and the sensitive group.The X~2 test was used,and P<0.05 was considered statistically significant.Results:(1)There was no significant difference in the occurrence of EMT between age and gender(both P>0.05).There was significant difference between the occurrence of EMT and the depth of invasion,malignancy and lymph node metastasis of gastric cancer(both P<0.05).(2)The positive rates of E-cadherin expression in the drug-resistant and sensitive groups were 18.2%and 60.9%,respectively,and the difference was statistically significant(P<0.05).The positive rates of N-cadherin expression in the resistant group and the sensitive group were 66.7%%and 21.7%,respec-tively,and the difference was statistically significant(P<0.05).The incidence of EMT(E-cadherin-negative,N-cadherin-positive)in the drug-resistant group was 54.5%;the incidence of EMT in the sensitive group was 13.0%.It can be seen that the incidence of EMT in the drug-resistant group was higher than that in the sensitive group.The difference between the drug-resistant group and the sensitive group was statistically significant(P<0.05).(3)The incidence of EMT in the experimental group A(patients with pos-itive CA72-4 and CA19-9,simultaneously)was 57.6%.In the experimental group B(except for patients with positive CA72-4 and CA19-9,simultaneous-ly),the incidence of EMT was 20.0%.Moreover,the difference between CA72-4 and CA19-9 expression and EMT was statistically significant(P<0.05).Conclusion:(1)In gastric cancer,the occurrence of EMT has a certain correlation with the depth of tumor invasion,malignancy and lymph node metastasis;The deeper the infiltration depth of gastric cancer occurs when EMT occurs,the worse the tissue differentiation,the higher the degree of malignancy,the more the number of metastatic lymph nodes.(2)The drug resistance of patients with gastric cancer before surgery is related to the occurrence of EMT,that is,gastric cancer patients with EMT(E-cadherin-negative and N-cadherin-positive)may develop drug resistance.(3)Patients with positive CA72-4 and CA19-9 are often related with the occurrence of EMT.It can be indirectly proved that the occurrence of EMT may be related to the drug resistance of gastric cancer... |
