Objective:This experiment study adopts MPTP induced Parkinson’s disease(hereinafter referred to as: PD)mouse model,through the climbing pole experiment,gait analysis,swimming experiment,field test,observation flavored gastrodia elata uncaria drink compared before and after the intervention in Parkinson’s disease model mice behavior,to verify that the flavored gastrodia elata uncaria drink can affect the behavior of Parkinson’s disease model mice,and to make preparation for the drug to the clinical.Methods: Subjects as male 60 C57 BL / 6 mice,were randomly divided into blank group,model group,flavored gastrodia uncaria decoction group,3 group of 20 points in each group 2,a total of 6 cage,intraperitoneal injection of saline blank group,model group and flavored gastrodia uncaria decoction group take 0.3% MPTP intraperitoneal injection of HCL solution,build subacute Parkinson’s disease mice model for seven days,after the success of the building,the blank group and model group to fill the stomach physiological saline for 15 days,flavored gastrodia uncaria decoction group give this drink 15 days,through field,swimming,climbing pole,gait analysis of the four experiments to test mice exercise behavior change.Results: 1.Climbing rod experiment: Model group climbing pole time significantly prolong than blank group(P < 0.01);In the three previous experiment,flavored gastrodia uncaria decoction mice climbing pole time compared with the model group was shorter(P < 0.05),statistically difference;As flavored gastrodia uncaria decoction lavage days increase,filling and feeding medicine 7,14 days,flavored gastrodia uncaria decoction group compared with model group climbing pole time significantly shortened(P < 0.01)2.Gait analysis: After the success of the model building,the average step model group was obviously shorter than the blank group(P < 0.01);Lavage for three days later,flavored gastrodia uncaria decoction group compared with model group,the average step length increased(P < 0.05);With the increase of gastric drug delivery days,since the beginning of the third experiment,flavored gastrodia uncaria decoction group average stepwas obviously longer than the model group(P < 0.01)..3.Swimming experiment:The first day of feed from irrigation to medicine,the model group compared with the blank group,model group mice floating obviously prolonged(P < 0.01);Via lavage for five days,flavored gastrodia uncaria decoction group compared with model group,flavored gastrodia uncaria decoction group floating time is shortened,to prolong swimming time(P < 0.05);With irrigation to increase in the number of times,flavored gastrodia uncaria decoction in the experimental group of mice with model group mice compared with 7,14 days(P < 0.01);Flavored gastrodia uncaria decoction group significantly prolong swimming time of mice.4.Mine experiment:Horizontal motion frequency model group was obviously lower than that of blank group(P < 0.01);Filling and feeding medicine 1,3 days,flavored gastrodia uncaria decoction group compared with model group,horizontal motion frequency increased(P < 0.05);Lavage for five days,flavored horizontal motion frequency in gastrodia uncaria decoction mice than mice in the model group increased significantly(P< 0.01);it has the extremely significant difference.Blank group vertical motion frequency compared with model group,model group significantly decreased(P < 0.01);Three days after lavage,flavored gastrodia uncaria decoction vertical movement more frequently than model group increased(P < 0.05);As the accumulation of dosing days in irrigation to the fifth day,the model group compared with flavored gastrodia uncaria decoction group,flavored gastrodia uncaria decoction group had a significantly increased frequency of vertical movement,it has the extremely significant difference;Last one experiment,the model group mice have a trend of recovery,but with flavored gastrodia uncaria decoction group compared with statistical difference(P < 0.05).Conclusion: 1.Normal C57BL/6 mice after MPTP intraperitoneal injection for 7 days after will generate static tremor,rigidity,less dynamic performance such as PD,explain MPTP possess neurotoxicity.2.Adding Tianma Gouteng Decoction can improve the exercise retardation,balance coordination,reduce the amplitude of tremor,improve the panic gaitand prolong the gait for MPTP-induced PD model mice.
