| Objective To explore the expression of ATP5 B protein in human breast cancer,and to analyze the differences in the expression of ATP5 B protein in breast cancer tissue and adjacent breast tissue.The correlation between the expression of ATP5 B protein in breast cancer tissues and the clinicopathological parameters including ER,PR,HER2,ki-67 and lymph node metastasis was statistically analyzed.The expression of ATP5 B in breast cancer cells was silenced to investigate the effect of ATP5 B on the biological function of breast cancer cells in vitro,and the possible mechanism of regulating ATP5 B protein expression and ectopic expression in tumor cells was preliminarily discussed.Methods Thirty samples of breast cancer tissue and adjacent breast tissue in our hospital were collected.The expression of ATP5 B protein was detected by immunohistochemistry.The correlation between the expression of ATP5 B protein in breast cancer tissues and the clinicopathological parameters including ER,PR,ki-67,HER2 and lymph node metastasis was statistically analyzed.To design ATP5 B siRNA plasmids.After the in vitro silencing of ATP5 B,the effects of ATP5 B on the biological function of breast cancer cells were investigated by cell scratch test,Transwell transfection test and MTT method.Results The expression of ATP5 B protein in breast cancer tissue was significantly higher than that in adjacent breast tissue(P < 0.05).There was no significant correlation between the high expression of ATP5 B protein in breast cancer tissues and the clinical and pathological parameters of ER,PR,ki-67,HER2 and lymphatic metastasis in breast cancer patients(P > 0.05).ATP5 B siRNA plasmid can significantly reduce the expression level of ATP5 B protein on T-47 D in breast cancer cells.In the cell scratch experiment,the motor ability of cancer cells in the ATP5 B silencing group was significantly lower than that in the control group(P < 0.05).Transwell assay showed that the migration ability of cancer cells in ATP5 B silencing group was significantly weaker than that in the control group(P < 0.05).MTT assay showed that the growth activity of cancer cells in the ATP5 B silencing group was significantly lower than that in the control group(P< 0.05).Conclusion The expression of ATP5 B protein in breast cancer tissue was significantly higher than that in adjacent breast tissue.The expression of ATP5 B protein in breast cancer tissue was not significantly correlated with the age,ER,PR,HER2,ki-67 and lymph node metastasis of breast cancer patients.Silencing the expression of ATP5 B protein in vitro can significantly inhibit the proliferation activity and migration ability of breast cancer cells. |
