Objective: To explore the relationship between the biological characteristics of ovarian cancer SKOV3 cell line before and after induction with celecoxib and artemisinin and epithelial-mesenchymal transition(EMT).Methods:(1)The ovarian cancer SKOV3 cell line was treated with celecoxib(10?M),and the EMT model of the cell line was established.The EMT model was induced by artemisinin(20 ?M,40 ?M,80 ?M),and the MET model of the cell line was established;(2)The proliferation of ovarian cancer SKOV3 cell line and EMT model cell line,ovarian cancer EMT model and MET model cell line were detected by CCK8 proliferation assay;(3)The ability of cell invasion and metastasis of ovarian cancer SKOV3 cell line and EMT model cell line,ovarian cancer EMT model and MET model cell line was detected by scratch test;(4)Western blot analysis was used to detect the difference in protein expression between epithelial calcium adhesion molecules and vimentin in ovarian cancer SKOV3 cell line and EMT model cell line,ovarian cancer EMT model and MET model cell line.(5)The expression of protein between ovarian cancer SKOV3 cell line and EMT model cell line,ovarian cancer EMT model and MET model cell line in epithelial calcium adhesion molecule and vimentin was detected by flow cytometry;(6)Immunofluorescence assay was used to detect the difference in protein expression between epithelial calcium adhesion molecules and vimentin in ovarian cancer SKOV3 cell line and EMT model cell line,ovarian cancer EMT model and MET model cell line.Results:(1)Compared with the parental ovarian cancer SKOV3 cell line,the EMT model cell line has enhanced proliferative ability in the same time period,and the MET modelcell line has weaker proliferative ability in the same time period than the EMT model cell line;(2)Compared with the parental ovarian cancer SKOV3 cell line,the EMT model cell line has the same ability of metastatic invasiveness in the same time.Compared with the EMT model cell line,the metastatic invasive ability of the MET model cell line is weakened in the same time;(3)Compared with the parental ovarian cancer SKOV3 cell line,the expression level of epithelial-associated protein(E-cadherin)was down-regulated in EMT model cell line,and the expression level of interstitial-associated protein(vimentin)was up-regulated compared with EMT model cell line.The expression level of epithelial-associated protein(E-cadherin)was up-regulated in the MET model cell line,and the expression level of the interstitial-associated protein(vimentin)was down-regulated;Conclusions:These results demonstrate that artemisinin has the ability to reverse celecoxib-induced epithelial-mesenchymal transition of SKOV3 cell lines.
