Objective:Guided by the basic theory of traditional Chinese medicine,the network pharmacology was constructed by using the drug gene target and metabolite component gene target in the database of traditional Chinese medicine.The potential mechanism of Fuzi Lizhong Pill on diabetic nephropathy was explored,which could provide guidance for clinical practice of syndrome differentiation and treatment of traditional Chinese medicine and seek the interaction between TCM prescriptions and syndromes.Methods:This study is divided into three parts.The first step is to search the components of Fuzi Lizhong pills from TCNID,TCM@taiwan,TCMSP and other traditional Chinese medicine databases,and then find out the predicted or confirmed gene targets of each component through stitcn,Swiss target predict ion and PubChcm datahascs.Secondly according to the metabolites detected by metabonomies din blood samples collected from patients wi th diabetic nephropathy in the pre-clinical stage of the resoarch group,the gone targets of metabol ites were identified through KEGG,Pubchem and other databases.Thirdly,using Cytoscape 3.5.1 software and Cluego plug-in,using topological analysis method,we enriched and analyzed the core part of Fuzi Lizhong pill gene target,the gene target of diabetic nephropathy metabolite,the same gene target of Fuzi Lizhong pill-diabetic nephropathy metabolite,constructed KEGG pathway and biological process network map,analyzed the interaction between networks,and discussed the attachment.The possible mechanism of Zilizhong Pill and diabetic nephropathy.Results:1.From TCMID,TCM@taiwan,TCMSP and other traditional Chinese medicine databases,we searched 1281 constituents in Fuzi Lizhong pills.According to the above compounds in stitch,Swiss target prediction and PubChem databases,a total of 1279 gene targets of Fuzi Lizhong pills were obtained.2.In our previous study,58 metabolites of diabetic nephropathy were detected,and 252 gene targets were identified by KEGG,Pubchem and other databases.3.After sorting out the above gene target results,KEGG pathways and biological process network maps of the core part of Fuzi Lizhong pill gene target,the metabolite gene target of diabetic nephropathy,the same part of Fuzi Lizhong pill-diabetic nephropathy metabolite gene target were constructed by introducing Cytoscape 3.5.1 software and plug-in.The results showed that the biological processes of Fuzi Lizhong Pill were mainly the transcriptional regulation of nucleic acid,chromosome and gene,the cellular response to hormones,compounds and inflammation,the modification of amino acids and proteins,and the KEGG pathway mainly included cancer,virus and transcription factor pathways.Metabolites of diabetic nephropathy are concentrated in the biological processes of amino acid metabolism,small molecule decomposition of organic matter,and horizontal transmission of neurotransmitters.KEGG pathway involves various metabolic pathways such as amino acids,nitrogen,fatty acids,nicotinic acid and nicotinamide.Their common biological processes include amino acid metabolism,neurotransmitter level regulation and synthesis,organic matter transport,drug decomposition,etc.Their common KEGG pathways are distributed in amino acid metabolism,glycolysis,drug metabolism,fatty acid metabolism,etc.Conclusion:Fuzi Lizhong Pill and diabetic nephropathy metabolite common gene target biological pathway,cell amino acid metabolism process P value is the smallest,in KEGG pathway also involves a number of amino acid pathways,suggesting that Fuzi Li zhong Pill may affect diabetic nephropauhy by interfering with the level of amino acid metabolism acid metaboli sm in intestinal environment.Through enrichment analysis,Fuzi Lizhong Pill may participate in the process of diabetic nephropathy by regulating fatty acid metabolism level,nucleotide transcription,improved antioxidant amino acid,neurotransmitter level,organic anion transport,synaptic transmission and small molecule metabolic pathway. |
