| [Background and Objective]Formaldehyde(FA)is a harmful gas,which induces neurotoxicity.Our research group has found that hydrogen sulfide(H2S),as a novel gas signal molecule,has definite antagonistic effect on the neurotoxicity of formaldehyde,but its mechanism has not been fully elucidated.Warburg effect is a metabolic mode of glycolysis in aerobic environment,which plays a role in regulating nervous function and is closely related to the occurrence and development of nervous system diseases.In this study,we first observed the effect of formaldehyde on Warburg effect in the hippocampus of rats,and then explored whether H2S revers this effect of formaldehyde.[Methods]1.Formaldehyde neurotoxicity model was established by injecting formaldehyde into lateral ventricle of male SD rats.2.Western Blot was used to detect the expression levels of Warburg effect-associated proteins in hippocampus:Hexokinase II(HKII),Pyruvate kinase M2(PKM2),Pyruvate dehydrogenase kinase 1(PDK1),Lactate dehydrogenase A(LDHA)and Pyruvate dehydrogenase(PDH).3.The content of lactate in hippocampal tissue was detected by lactic acid kit.[Results]1.Formaldehyde upreglulated Warburg effect in the hippocampus of rats.After treatment with FA(0.1,1,10μmol)for 7 d,the expressions of Warburg effect-associated proteins HKII,PKM2,PDK-1 and LDHA in hippocampus were increased,the expression of PDH was decreased,and the content of lactate was increased,indicating that the Warburg effect was enhanced in the hippocampal tissue of rats treated with formaldehyde.2.H2S inhibited formaldehyde-induced Warburg effect in the hippocampus of rats.After pretreatment with Sodium hydrosulfide(NaHS,a donor of H2S,10,30,100μmol/kg,i.p.),the expressions of Warburg effect-related proteins HKII,PKM2,PDK1 and LDHA were decreased,the expression of PDH protein was increased,and the content of lactate was decreased in the hippocampus of rats exposed to formaldehyde.These results indicated that H2S inhibits formaldehyde-induced upregulation in hippocampal Warburg effect.[Conclusion]Hydrogen sulfide inhibits Warburg effect in the hippocampus of rats exposed to formaldehyde. |
