Obiective:To compare the differences in the histological characteristics between the loose bodies(LBs)of synovial chondromatosis(SC)and normal cartilage.To explore the function of the Interleukin-6(IL-6)in the developing of the SC.Method:Ten SC LBs and ten normal LBs from injured knee with exfoliation of the articular cartilages were collected,all of the LBs were made into paraffin sectons and the hematoxylin-eosin staining(HE)was took.The content of aggrecan and collagenⅡwas accessed through the Safranine O staining and IHC,respectively.Then the average optical density was measured through the Image Pro Plus(IPP)6.0.The positive cell’s percentage was calculated after the IHC.The difference between the two kinds of the LBs was assessed.The synovium was collected from the bursa suprapatellaris of the patients undergoing knee arthroscopy surgery,the synoviocytes were dissociated and cultured to the passege 3th.The five experimental teams including team A,team B,team C,team D and team E were prepared and the H-DMEM containing 10%FBS absence of IL-6+soluble IL-6receptor(sIL-6R)or in presence of 100ng/mlIL-6,100ng/mlsIL-6R,50ng/ml IL-6+sIL-6R,100ng/mlIL-6+sIL-6R was used,respectly.The control groups included team A,team B,team C and the experimental groups included team D,team E,every team had six samples.Three samples from the every teams were took out and the immunofluorescence technique(IF)was performed to investigate the protein expression level of collagenⅡ,the intensity of IF was accessed through the IPP6.0 and the compare was took both in intragroups and among groups.Results:After HE staining,The distribution of the chondrocytes was found regularly in the cartilaginous tissue of the normal LBs but not for the SC LBs and there were much more hyaline chondrocytes in the SC LBs than that of the normal LBs.The average optical density after the Safranine O staining was(0.069±0.014)and(0.141±0.014)for SC LBs and normal LBs,respectively.The average optical density after the IHC was(0.035±0.009)and(0.017±0.007)for SC LBs and normal LBs,respectively.The average positive cell’s percentage of IL-6 was(88.07%±4.16%)and(21.06±7.50%)for the SC LBs and normal LBs,respectively.The intensity of immunofluorescence was(21.40±1.03),(21.78±0.89),(22.32±0.36),(30.11±0.99),(37.39±0.67)in the7days’five groups,respectly and was(22.61±0.64),(22.08±0.96),(22.20±0.70),(38.23±0.78),(44.86±1.12)in the 14days’five groups,respectly.There were no statistic difference on the mean fluorescence intensity in the group A,group B,group C(P>0.05),but for the group D,group E(P<0.05),and there were statistic difference between group D,group E and group A,group B,group C both in intragroups and among groups(P<0.05).The mean fluorescence intensity of the 14days was much more than that of the 7 days,not only for the group D but also for the group E(P<0.05).Conclusions:The SC LBs has the similar histological characteristics to the normal cartilage.IL-6 was highly expressed in the SC LBs and may contribute to hyaline chondrogenic differentiation of human synoviocyte.
