Objective:To investigate the effect of resveratrol on the proliferation of gastric cancer cell line MKN45 and its effect on MEK/ERK signaling pathway,and to provide data and ideas for clinical treatment and new drug development of gastric cancer.Methods:Cells were treated with resveratrol at doses of 0,100 and 400μmol/L,while the inhibitor of MEK/ERK pathway,PD98059(20mg/L,100μL)was used as positive group.After treatment of each group of cells for 4 hours.MTT assay was used to analyze the inhibition rate of cell proliferation.Western blotting was used to analyze the expression of apoptotic related proteins,including Bax,Bcl2,Caspase3,Caspase9 and MEK/ERK pathway-associated proteins,including Ras,Raf,MEK,ERK1/2.Cellular immunofluorescence was used to analyze the fluorescence intensity of MEK and ERK1/2.Results:1.MTT assay showed that: The results found that compared with the control group,the proliferation of cells in PD98059 group,low dose resveratrol group and high dose resveratrol group was significantly inhibited after treatment for 1h(P<0.05),2 h(P<0.05)and 4 h(P<0.05).2.Western blotting showed that :Compared with the control group,the expressions of Bax,Caspase3 and Caspase9 in PD98059 group,low dose resveratrol group and high dose resveratrol group were significantly increased(P<0.05).The expression of Bcl2,Ras,Raf,MEK and ERK1/2 were significantly decreased(P<0.05),the difference was statistically significant.These results indicated that resveratrol induced a significant concentration-and time-dependent inhibitory effect on cell proliferation.3.Cellular immunofluorescence assay showed that :When immunofluorescence chemistry was used to detect changes in MEK/ERK signaling pathway protein levels,MEK and ERK1/2 proteins were also found to be mainly localized in the cytoplasm.The fluorescence intensity of ERK1/2 in PD98059 group,low dose resveratrol group and high dose resveratrol group were significantly decreased.Conclusions:1.Resveratrol induces a significant concentration-and timedependent inhibitory effect on the proliferation of gastric cancer cells.2.Resveratrol induces apoptosis of gastric cancer cell line MKN45.3.The inhibitory effect of resveratrol on gastric cancer cells may be related to the MEK/ERK signaling pathway.
