Rhamnus utilis is a deciduous shrub or small tree.Its fruit can be used folk medicinal for antipyretic,liver protection,and anti-diarrhoea effects.At present,due to the lack of the theoretical foundation about the active components of R.utilis fruit,the important medical value is not large-scale exploited.In this study,R.utilis fruits were used as material to research the general chemical constituents and ultrasonic extraction technology of flavonoids.Optimized macroporous resin purification process by response surface method.The effect of flavonoids from R.utilis fruits on acute liver injury induced by CCl4 was carried out in ICR mice.This study was aimed to provide reliable experimental basis for the treatment or prevention of acute liver injury.The main contents and results of the study are as follows:1.The chemical constituents of R.utilis fruit and leaves were extracted by Water,ethanol and petroleum ether from R.utilis fruit,and chemical changes such as color change,precipitation or not were used to determine the effect of extracting solution on the growth of R.utilis fruit.It was concluded that both fruit and leaves contained polysaccharides,organic acids,phenols,tannins,flavonoids,steroids,triterpenoids,volatile oils,and may contain anthraquinones and cardiac glycosides.R.utilis fruit contains coumarin,lactone,grease,and leaves do not contain these substances.2.The most suitable macroporous resins,AB-8,was selected by static adsorption-desorption test from six macroporous resins with different polarity.The single factor experiment of dynamic adsorption and desorption,combined with response surface method to optimize the technological conditions of separation and purification of R.utilis fruit flavonoids.The optimal purification conditions were loading 2 BV of 0.3 mg/mL flavonoids with pH 3at a flow rate of 4 mL/min,and subsequently eluting the column with 4 BV of80%ethanol at a flow rate of 4.5 mL/min.The ratio of absorption,desorption and recovery of flavonoids in R.utilis fruits were(85.6±0.30)%,(71.26±0.28)%and(94.23±0.40)%,respectively.3.The effect of R.utilis fruit flavonoids on acute chemically damaged liver was explored on the CCl4 acute liver injury model in ICR mice.The results showed that,Middle-(100 mg/mL)and high-dosage(200mg/mL)flavonoids could significantly decrease the content of ALT,AST in serum and the content of MDA in liver,and significantly increase the content of SOD,CAT in liver.It showed the same pharmacological direction as the biphenyl diester,which has a protective effect on the liver.The results indicated that flavonoids in R.utilis fruit at a certain dose could protect the acute liver injury induced by CCl4 in mice.
