Objective:To survey the expression level of Twist2 in human glioma cell line;to explore whether the possible mechanism of Twist2 affecting invasion and migration of glioma is an epithelial-mesenchymal transition(EMT).Methods:1.Using the methods of q-PCR to detect the expression of Twist2 mRNA in human glioma cell lines U87,T98 G,H4,U118 and SHG44.2.Overexpression of the Twist2 gene plasmid lentivirus infected human glioma cell lines U87 and T98 G.Each group of cell lines was divided into three groups: the first group: blank group(Blank);the second group: overexpression negative control group(NC-ox);the third group: overexpressed Twist2 genome(Twist2-ox).3.Using the methods of CCK-8 method,Transwell chamber invasive test and Wound healing scratch test to detect cell proliferation,invasion and migration respectively.4.Nude Mice were tested in vivo to test the invasion and migration ability.5.Western blot was used to detect the expression levels of EMT-related proteins: E-cadherin,N-cadherin,Vimentin and Fibronectin.Resμlts:1.The expression of Twist2 gene mRNA was expressed in human glioma cell lines U87,T98 G,H4,U118 and SHG44,and the expression level in U87 and T98 G was relatively low.2.CCK-8 method showed that there was no significant change in the proliferation of Twist2 genomic cells.The difference was not statistically significant.Transwell chamber invasion and Wound healing scratch test showed that the invasion and migration ability of Twist2 group was significantly enhanced.Learning meaning.3.In vivo nude mice tumor formation experiment,overexpression of Twist2 genome in the tumor volume is larger,faster growth and the difference is statistically significant.4.Overexpression of Twist2 genome U87 and T98 G EMT-related western blot assay protein: Overexpression of Twist2 group E-cadherin expression was significantly decreased,the difference was statistically significant;and N-cadherin(N The expression levels of-cadherin,vimentin and fibronectin increased significantly,and the difference was statistically significant.Conclusions:The The possible mechanism by which Twist2 promotes the invasion and migration of human glioma cell lines U87 and T98 G is an EMT.
