Objective: To observe the correlation of serum triglyceride and other insexes with oxidative stress levels in different lipid-resistant population after high fat meal load.Methods: There are 109 volunteers recruited in Hebei Provincial People’s Hospital.All volunteers filled out questionnaires and signed informed consent form,to carry out the high-fat diet experiment.The indexes such as height and weight were recorded.Carotid intima thickness was detected by ultrasonic method.The fasting blood were collected at 0h,2h,4h,6h respectively after oral fat load.The blood samples were used to detect triglycerides(TG),fasting plasma glucose(FPG),fasting insulin(FINS),total cholesterol(TC)and other indicators.Serum was stored in-80 ℃ refrigerator preservation.Oxidative stress indicators including super-oxide dismutase(SOD),and malondialdehyde(MDA)were determinated.According to serum TG level on fasting and 4 hour TG after meal,the volunteers were divided into three groups: the normal fat tolerance group(NFT),impaired fat tolerance group(IFT)and hyper-triglyceridemia group(HTG).The basic indexes such as BMI,WHtR,FPG and FINS were evaluated by variance analysis.The variance analysis was used to evaluate the dynamic changes and differences of each group with time,TG,MDA,SOD and other indexes.The correlation of SOD with TG was evaluated by unary linear regression analysis.The association between SOD and TG,MDA,HOMA-IR,HOMA-β,BMI and WHtR was analyzed by multivariate linear stepwise regression.Results:1 The data of the general index of each group of volunteers were analyzed.The results showed that there were no statistical differences in gender among the three groups.The NFT group was younger than the other two groups,the difference was statistically significant(P<0.05),and there was no statistical difference between the two groups of IFT and HTG,(P>0.05);BMI,WHtR,FPG,FINS and HOMA-IR in three groups with the decrease of lipid tolerance level grstatisticaladually increased,the difference was statistically significant(P<0.05).There was no significant differennce in HTG and IFT group(P>0.05),HOMA-β in NFT group was lower than the other two groups with significant difference.There were statistically significant difference in fasting AST/ALT between the two groups of NFT and HTG(P<0.05).There was no statistically significant difference between IFT and the other two groups(P>0.05).The comparison between fasting ApoA1/ApoB and the other two groups was statistically significant,while there was no statistically significant difference between the IFT and HTG(P<0.05).2 There was 1 case(2.63%)of NAFLD in NFT group,11 cases of NAFLD in IFT group(28.95%),19 cases of NAFLD in HTG group(57.58%): The difference was statistically significant(P<0.05).The thickness of intima-media thickness(IMTc)in each group increased with the decrease of lipid tolerance,but the difference was not statistically significant(P>0.05).3 The lipid metabolism indexes of each group(1)TG level in three groups showed an upward trend.TG of HTG group and IFT group went on peak at 4h.At each point TG showed a trend as follows: HTG group > IFT group > NFT group;the difference was statistically significant(P < 0.05).The contrast test in the main body of repeated measurement variance analysis showed that there were significant differences in TG in the three groups at different time points,and therewere significant differences of TG between the three groups(P<0.05).(2)TC level in the three groups showed an upward trend,at each point the TC level showed the trend as follows: HTG group>IFT group>NFT group,the difference was statistically significant(P < 0.05).Repeated measurement variance analysis showed that there were significant differences among the three groups of TC levels at each time point,and there were significant differences among the three groups(P<0.05).(3)HDL-C level in three groups showed an decrease trend.The HDL-C levels in three groups showed the trend as follows: NFT group>IFT group>HTG group.Repeated measurement variance analysis showed that the HDL-C level in three groups were significantly different(P<0.05).4 The oxidative stress indexes of each group(1)After a high-fat diet,the serum MDA in three groups of volunteers showed an overall continuous upward trend,and HTG group>IFT group> NFT group.The analysis of the variance of repeated measurements shows that there were significant differences in the three groups at different points of time after eating high-fat diet(P<0.05).The inter-subjectivity effect test showed that there were differences in the HDL-C levels of the three groups(P<0.05).(2)The serum SOD in the three groups showed an overall continuous decline trend and the NFT group>IFT group>HTG group.However,the SOD in IFT group had a slight increase trend of SOD from 4h to 6h.There were significant differences of SOD at different points after eating high-fat diet(P < 0.05).There were significant differences of SOD levels in the three groups(P<0.05).5 The effects of BMI,WHtR,HOMA-IR,HOMA-β,TG4 h and MDA4 h on SOD were observed by multiple linear stepwise regression analysis.The results showed that TG,MDA4 h and HOMA-IR were important impact factors of SOD(P < 0.05),while BMI,WHtR and HOMA-β had no significant effect on SOD(P>0.05).Conclusion:1 After oral high-fat diet test,with the increase of TG,the body oxidative stress level increased,the antioxidant capacity decreased.2 After fasting and high fat diet serum TG level was closely related to the increase of insulin resistance.3 The more lipid tolerance decreased,the higher the prevalence of NAFLD.
