Objective:To investigate the effect of AngⅡ on the expression of periostin in foam cells and its possible mechanism.Methods:1.RAW264.7 was cultured in vitro and induced by ox-LDL(50mg/L)into foam cells.The foam cells were randomly divided into groups:(1)different concentration groups: AngⅡ intervention with concentrations of 0nmol/L,10nmol/L,100nmol/L,1000nmol/L and 10000nmol/L was added for 24h;(2)different time groups: AngⅡ intervention with a concentration of 1000nmol/L was added for 0h,3h,6h,12 h and 24 h respectively.2.The foam cells were randomly divided into four groups:(1)control group: no intervention;(2)AngⅡ group: 1000nmol/L AngⅡ;(3)BAY11-7082(NF-κB inhibitors)+AngⅡ group: BAY11-7082(1h)+1000nmol/L AngⅡ(24h);(4)BAY11-7082 group:BAY11-7082(1h).3.The expression of periostin mRNA in each group was detected by RT-PCR.The expression of periostin protein and phosphorylated NF-κB p65 protein in each group was detected by Western Blotting.Results:1.AngⅡ promotes the expression of periostin in foam cells:(1)at the same time,periostin mRNA and protein expression increased with the increase of AngⅡ concentration(P<0.05).When the concentration reached 1000nmol/L,periostinexpression level was the highest.(2)with the same concentration,periostin mRNA and protein expression increased gradually with the extension of AngⅡ action time(P<0.05),reaching the peak at 24 h.2.AngⅡ regulates the expression of periostin through NF-κB signaling pathway:AngⅡ promotes the expressions of phosphorylated NF-κB p65,periostin protein and periostin mRNA in foam cells.Compared with AngⅡ group,the expressions of phosphorylated NF-κB p65,periostin protein and periostin mRNA were decreased after the addition of BAY11-7082.Conclusion:1.AngⅡ promotes the expression of periostin in foam cells with concentration and time dependence.2.AngⅡ regulates periostin expression through NF-κB signaling pathway.
