Objective: We aim to establish obese mice models by means of high-fat diet,and spontaneously constructed ob-/-,db-/-mice models,and to investigate the morphological structure and associated molecular biomarker of PDGs induced by several obese mice models.Methods:Establishment and grouping of obese mice models:Group HFD(n=10):Construction of obese mice models in C57BL/6 through high-fat diet for 3 months;Group HFD-ctrl(n=6):A regular diet for C57BL/6 mice in the same period for 3 months;Group OB(n=8):The ob-/-mice of 7-8 weeks receive a regular diet for 4 weeks;Group OB-ctrl(n=6):The ob-/w mice of 7-8 weeks receive a regular diet for 4 weeks;Group DB(n=8):A regular diet for db-/-mice of 7-8 weeks for 4 weeks;Group DB-ctrl(n=6):A regular diet for db-/w mice of 7-8 weeks for 4 weeks.Weight,the glucose,triglyceride and total cholesterol are measured.The structure of PDGs in mice and the effect of obesity on the structure and quantity of PDGs are identified by HE(hematoxylin-eosin staning).The expression of TFF2 and MUC5AC are analyzed by IHC,the change in staining of Alcian blue in PDGs are evaluated.Results:Weight,glucose,triglyceride and total cholesterol in obese mice are significantly higher than the control groups.PDGs resides within the mesenchyme surrounding larger pancreatic duct,the number of PDGs in obese mice are increased compared with the lean mice group.TFF2 and Alcian blue are specific markers of PDGs,the expression of TFF2 are up-regulated and the staining of Alcian blue increases in obese mice,the expression of MUC5 AC is negativedownregulated? in all mice.Conclusion: The models of obese mice via high-fat diet are successfully established and obesity promotes the proliferation of PDGs,and facilitates the secretion of acid mucin and enhances the expression of TFF2,which may be related to the repair of pancreatic duct injury.
