Tinglishengmai Prescription is the experience prescription which is treating Congestive Heart Failure summarized by Xing Yuepeng,a famous veteran TCM doctor in China,based on many years of clinical experience.After clinical verification,it can effectively alleviate the symptoms of patients,improve the quality of life and reduce hospitalization time.In order to make this prescription more widely used in clinic,we modified the prescription to prepare oral liquid,and studied the extraction process and fingerprint of the oral liquid.Objective:To maximize the retention of pharmacodynamic components,response surface methodology(RSM)and information entropy method(IEM)were used to determine the optimum ethanol extraction process parameters of Tinglishengmai oral liquid.In order to ensure the quality,safety and stability of the oral liquid,the fingerprint analysis method of the oral liquid was established by high performance liquid chromatography(HPLC),which can provide a method for the quality evaluation of the oral liquid.Methods:The optimum alcohol extraction process of Tinglishengmai Oral Liquid:Based on single factor experiment,taking extraction time,material-liquid ratio and alcohol concentration as influencing factors,the content of ginsenoside Re,ginsenoside Rg1,salvianolic acid B,ligustrazine,schisandrin A and alizarin B-23 acetate as evaluation indexes,the best alcohol extraction process was screened by response surface method combined with information entropy method.。Establishment of fingerprint of Tinglishengmai Oral Liquid:The chromatographic column YMC J’sphere ODS-H80-C18(4.6 mm*250 mm,4micron)was used to detect the fingerprint of Tinglishengmai oral liquid.The mobile phase used was acetonitrile in channel A and phosphoric acid in channel B with gradient elution.The flow rate was determined to be 1.0mL/min.The detection wavelength was 203 nm,270 nm and the column temperature was 30 C.The fingerprint similarity of 10 batches of different oral liquefaction components was calculated by evaluation software.Results:The technological parameters of ethanol extraction of Tinglishengmai oral liquid were as follows:extraction time 3.5 h,ratio of material to liquid 1:10,alcohol concentration 82%.The established chromatographic conditions can make the components of Tinglishengmai oral liquid well separated.The similarity between 10 batches of samples is more than 0.98.There are 21 common peaks at 203 nm and 17 common peaks at270 nm.Conclusions:By using response surface methodology and information entropy method,the optimum ethanol extraction process parameters are stable and reliable,which can provide experimental basis for the development of Tinglishengmai Prescription and modern Chinese medicine preparation.The established fingerprint analysis method has good precision,repeatability,stability and high separation between common peaks.It can be used for comprehensive evaluation of the quality of Tinglishengmai oral liquid.
