| Objective:In this experiment,the effects of huoxue tongluo qingre lishi precription on the expression of platelet source growth factor(PDGF),vascular endothelial growth factor(VEGF)and receptor Flk-1 in IgA nephrotic rats were observed.In order to explore the possible mechanism of treating IgAN rats with huoxue tongluo qingre lishi precription,it provides experimental theoretical basis for the clinical use of huoxue tongluo qingre lishi precription to treat IgAN.Methods: 40 healthy and clean male SD rats were tested negative after 1week of adaptive rearing,and were randomly split up into 4 groups: normal group,model group,beinapuli group and traditional Chinese medicine group.The method of bovine serum albumin + lipopolysaccharide + carbon tetrachloride + castor oil was used to replicate the model of IgA nephropathy in rats.After the successful replication of the model,the normal group and the model group were given distilled water by gavage,beinapuli group was given beinapuli by gavage,and the Chinese medicine group was given the chinese herbal compound by gavage once a day for 4 weeks.During the experiment,rats were regularly tested for urine analysis and 24-hour urinary protein quantification.The rats were sacrificed at the 14 th week,and serum and renal tissues were collected for the detection of the serum creatinine(Scr),urea nitrogen(BUN),serum total protein(TP),total cholesterol(TC),high density quality protein(HDL),and other biochemical indicators,glomerular pathological changes,and the expression of platelet derived growth factor(PDGF),vascular endothelial growth factor(VEGF)and receptor flk-1 in renal tissues.Result:1.The rats in the normal group had a good general condition during the entire experimental process;At the beginning,the other three groups of rats gradually showed varying degrees of activity reduction or restlessness,mental disorders,irritability,sparse hair,and decreased feeding.Given the corresponding treatment,the general status of rats in beinapuli group and traditional Chinese medicine group improved compared with the model group.2.The serum creatinine(Scr),urea nitrogen(BUN),serum total protein(TP),total cholesterol(TC)and high density quality protein(HDL)of 4 groups of rats were not statistically significant(P>0.05).3.There was no proteinuria during the experimental period in the normal group of rats,and the 24-hour urinary protein in the other 3 groups was significantly increased.After the corresponding treatment,the rats in the Beinapuli group and the traditional Chinese medicine group significantly decreased compared with the model group(P<0.05).There was no statistical significance compared with the traditional Chinese medicine group and the beinapuli group(P>0.05).4.There were no pathological changes of kidneys in normal rats,and the pathological changes of kidneys were heavier in the model group than in the beinapuli group and the traditional Chinese medicine group.5.The expressions of platelet source growth factor(PDGF),vascular endothelial growth factor(VEGF)and receptor Flk-1 in the renal tissue of rats in model group,beinapuli group,and traditional Chinese medicine group was significantly increased compared with normal group(P<0.05);The expression of beinapuli group and traditional Chinese medicine group was significantly reduced compared with the model group(P<0.05);beinapuli group and traditional Chinese medicine group had no statistical significance(P>0.05)Conclusion:1.The 24-hour urinary protein quantification of the rats with Ig AN can besignificantly reduced by huoxue tongluo qingre lishi precription that can be protect kidney function.And its effect is comparable to that of beinapuli.2.The pathologic damage of kidney tissue in rats with IgAN can be significantly reduced by huoxue tongluo qingre lishi precription,with the progress of the disease can be delayed.And its effect is comparable to that of beinapuli.3.The expression of PDGF,VEGF and receptor Flk-1 in the kidney tissue of IgA nephrotic rats can be significantly reduced by huoxue tongluo qingre lishi precription,inhibiting mesangial cell proliferation,reducing extracellular matrix synthesis,maintaining glomerular basement membrane stability,improving the renal pathological changes,slowing the development of the disease.And its effect is comparable to that of beinapuli. |
