| Objective: On the basis of the previous research of the research group,separated and purified the chemical constituents of the Mongolian medicine Sendeng-4,study on the pharmacodynamic components of Mongolian medicine compound Sendeng-4 in vivo.The anti-inflammatory and analgesic effects of Mongolian medicine Sendeng-4 on mice were studied by granuloma test,ear swelling test and writhing reaction test.the hyperuricemia model of rats was established with potassium oxalate to study the hypouric acid effect of Sendeng-4 on rats.The contents of total flavonoids and terpenoids in serum of Sendeng-4healthy rats and rheumatoid arthritis rats were studied by ultraviolet-visible spectrophotometry.And explore its migration in serum.The contents of total flavonoids and terpenoids in the metabolism of Sendeng-4 healthy rats and rheumatoid arthritis rats were studied,therapeutic effects of total flavonoids and terpenoids on rheumatoid arthritis in rats,The contents of genipin-1-O-β-D-gentinoside and Geniposide in joints of healthy rats,rheumatoid arthritis rats and hyperuricemia rats were determined by UHPLC-TOF/MS,the time curve was also studied,to provide the basis for the pharmacological and pharmacodynamic study of Mongolian medicine compound Sendeng-4.Method: The Mongolian medicine Sendeng-4 was made into a extract,the upper hole was adsorbed to the resin column,concentrated and dried,and the site 2 was obtained.The site 2was continued with the large hole resin column,silica gel column,MCI column,and gel column.Wash off,get different flow points,repeatedly wash and purif.Targeted tracing of myricetin,dihydromyricetin,quercetin,gardenoside,genipin-1-O-β-D-gentinoside and gallic acid was carried out.180 mice were taken for cotton granuloma-induced granuloma experiments,xyleneinduced mouse ear swelling experiments,and acetic acid-induced mouse writhing reaction experiments.The difference in granuloma weight gain between the Mongolian medicine Sendeng-4 administration group and the model blank group and the positive control group,the difference in the inhibition of ear swelling and the writhing inhibition in mice were observed.Rats were randomly divided into normal group and hyperuricemia model group,and uric acid(UA)levels in serum and urine of each group were determined by ELISA.Rats were divided into healthy group and rheumatoid arthritis model group,oral administration of Mongolian medicine Sendeng-4 effective part.After administration,rat urine and serum at different time points were taken.After protein removal and colour reaction,determination of total flavonoids and terpenoids in rat serum and urine by Ultraviolet Spectrophotometry,calculate the content,serum pharmacokinetics was also studied.Rats were divided into normal group,rheumatoid arthritis model group and hyperuricemia model group.After intragastric administration of Mongolian medicine Sendeng-4 effective part,take the acetonitrile extract from the rat joint,UPLC-Q-TOF-MS was used to determine genipin-1-O-β-D-gentinoside and Geniposide in the extract.Result: Six compounds were isolated and enriched from the Mongolian medicine Sendeng-4.After identification and comparison with the analysis of the nuclear magnetic spectrum,it was confirmed as bayberry,dihydrobayberry,gardenosides,and Jingniping-O-β-D-gentian diglycosides,quercetin,Gallic acid.The Mongolian Medicine Compound Sendeng-4 middle dose group(135 mg/kg)can inhibit the growth of mouse granuloma induced by cotton balls,inhibit the xylene-induced ear swelling in mice,and inhibit the writhing reaction induced by glacial acetic acid in mice.It shows that Mongolian medicine compound Sendeng-4 has significant anti-inflammatory and analgesic effects.UA levels in serum and urine of rats were measured and compared between model control group and drug group.The results showed that Mongolian medicine Sendeng-4 had the effect of reducing uric acid.A method for the determination of total flavonoids and total strontium in rat serum by UV-visible spectrophotometry was established.The pharmacokinetics of total flavonoids and total quinones were studied.The content of genipin-1-O-β-D-gentiobioside and geniposide in rat joints was determined by UPLC-Q-TOF-MS method.Conclusion:Mongolian medicine compound Sendeng-4 has significant anti-inflammatory and analgesic effects,and can reduce the level of uric acid,which corresponds to its clinicaluse in the treatment of rheumatoid arthritis and gout.Studies on the pharmacodynamic components of Mongolian medicine Sendeng-4 showed that the total flavonoids and total sputum contained in the compound had important significance in the treatment of rheumatoid arthritis,verification of terpenoid quality markers: genipin-1-O-β-D-gentinoside and geniposide by UPLC-Q-TOF-MS,it provides a powerful basis for the pharmacological and pharmacodynamic study of Mongolian medicine Sendeng-4. |
