Objective: In this study,we aimed to ? evaluate of the value of seminal plasma miR-210 as a potential biomarker in screening patients with impaired spermatogenic function leading by varicocele;?explore the mechanism of miR-210 in the impairment of spermatogenic function caused by varicocele.Methods: ? Evaluation of the value of seminal plasma miR-210 as a potential biomarker in screening patients with impaired spermatogenic function leading by varicocele Semen parameters and miR-210 expression levels of semen samples were detected by semen analysis technique and quantitative real-time polymerase chain reaction(q RT-PCR)technique,respectively.Cox univariate and multivariate analysis and receiver operating characteristic(ROC)curves were used to evaluate the ability of seminal plasma miR-210 to screen patients with impaired spermatogenic function leading by varicocele.? Exploring the mechanism of miR-210 in the impairment of spermatogenic function caused by varicocele The expression of miR-210 in mouse spermatogonia(GC-2)was detected by q RT-PCR.The cell transfection technique was used to up-regulate and down-regulate miR-210 in GC-2 cells,and the CCK8 assay was used to detect the change of cell proliferation rate after GC-2 cells were up-regulated/down-regulated.The mechanism and association of miR-210 and Caspase-3 in the impairment of spermatogenic function caused by varicocele was verified by Western Blot.Results: ? miR-210 was stably expressed in seminal plasma of patients with varicocele and healthy males,and had the characteristics as a biomarker.The expression of miR-210 in seminal plasma of patients with varicocele was significantly higher than that of healthy males(p < 0.01).The expression of miR-210 in seminal plasma of patients with varicocele was positively correlated with the severity of varicocele,and its expression increased with the severity of the disease.The expression of miR-210 in seminal plasma of varicocele patients with impaired spermatogenesis was significantly higher than that of spermatogenic function was not damaged(p < 0.01).The ROC curve showed that seminal plasma miR-210 had significant value in screening patients with impaired spermatogenesis caused by varicocele: AUC=0.94,p < 0.0001;sensitivity = 88%,specificity = 90%.? The expression level of miR-210 in GC-2 cells was stable.After upregulation of miR-210 in GC-2 cells,the proliferation of GC-2 cells decreased significantly;conversely,the proliferation of GC-2 cells was significantly increased after downregulation of miR-210 in GC-2 cells.There was no significant difference in the expression level of Pro-caspase-3 between each group of GC-2 cells;the expression of Cleaved caspase-3 in the up-regulation group was significantly increased,while that was significantly reduced in the down-regulation group.Conclusions: ? The expression of miR-210 was stable in varicocele and healthy male seminal plasma,and had the characteristics of becoming a biomarker;? The expression of miR-210 in seminal plasma of patients with varicocele was significantly higher than that of healthy males,and its expression increased with the severity of varicocele;? The expression level of seminal plasma miR-210 can be reversed to the level of healthy males? after surgery;? Seminal plasma miR-210 can be used as a biomarker to screen patients with impaired spermatogenesis caused by varicocele;? We found that miR-210 can induce the apoptosis of spermatogenic cells by promoting the activation of Caspase-3.
