Objective:Treatment of acute myocardial infarction rats with rhG-CSF.The relationship between Angiopoietin 2 and CD105 and the infarct size of AMI rats after interventing rhG-CSF was investigated by detecting the expression of Angiopoietin 2and CD105 genes in the myocardium of acute myocardial infarction rats and the infarct size of acute myocardial infarction rat model.Methods:There were sixty healthy and clean SD rats aged 6-8 weeks and weighing200-300 g,They were randomly divided into three experimental groups:rhG-CSF group,sham-operated group,(AMI group)acute myocardial infarction group.And every group was randomly divided into two groups,one week group and two weeks group,with 10 rats in each group.(1)Sham-operated group: The left anterior descending branch of rats was threaded without ligation after thoracotomy,and 0.9%sodium chloride injection was intraperitoneally injected 24 hours after operation for5 consecutive days.(2)AMI group: Acute myocardial infarction model was induced by ligation of left anterior descending coronary artery in rats,24 hours after operation,intraperitoneal injection of 0.9% sodium chloride injection for 5consecutive days;(3)rhG-CSF group: Acute myocardial infarction model was induced by ligation of left anterior descending coronary artery in rats,24 hours after operation,intraperitoneal injection of rhG-CSF for 5 consecutive days(10ug/kg/d).Penicillin was injected intraperitoneally into each group for 3 consecutive days.On the 7th and 14 th day after anesthesia,the rat hearts were taken.Detecting the expression of Ang2 and CD105 in myocardium by RT-PCR,and the infarct size ofacute myocardial infarction rats was calculated by TTC staining.Results:(1)Ang2 test results showed that there was no significant difference in Ang2 gene expression between the one week and the two weeks group in sham operation group(P > 0.05);in the AMI group,the gene expression level of the two weeks group was higher than that of the one week group(P < 0.01),and higher than that of the sham operation group(P < 0.05);in the intervention group,the gene expression level of the two weeks group was higher than that of the one week group(P < 0.01),and higher than that of the same period AMI group(P < 0.05).(2)The results of CD105 test showed that there was no significant difference in the gene expression level of CD105 between one week group and two weeks group in sham operation group(P > 0.05);in AMI group,the gene expression level of two weeks group was higher than that of one week group(P <0.01),and higher than that of sham operation group(P < 0.01).In intervention group,the gene expression level of two weeks group and one week group was higher than that of the same period AMI group(P < 0.01),but there was no statistical significance difference between two weeks group and one week group itself(P > 0.05).(3)The results of myocardial infarction area test showed that in the AMI group and the intervention group,the area of myocardial infarction in the two-week group was less than that in the one-week group(P < 0.01),and the area of myocardial infarction in the intervention group was less than that of the same period AMI group(P < 0.01).Conclusion:(1)The gene expression levels of Ang2 and CD105 in myocardium of rats with acute myocardial infarction were increased in 2-week group than in 1-week group.(2)After rhG-CSF intervention,the expression of Ang2 and CD105 in myocardial tissue of rats with acute myocardial infarction was further increased in each time group,which was related to the reduction of infarct size.
