Objective: Tumor metastasis is the main cause of death in patients with colorectal cancer(CRC).However,research on finding m RNA expression profiles for predicting colorectal liver metastases(CLM)with bioinformatics remains limited.Methods: All the statistical analysis was performed with Robust Multichip Average(RMA).Using a m RNA mining approach,we investigated the m RNA expression profiling in CRC cohorts from the Gene Expression Omnibus(GEO).We identified CLM-related m RNAs in two datasets(GSE81980,GSE18105).By random forest,twelve gene signatures(differential genes)were further selected to develop the risk biomarkers of CLM,which were significantly associated with metastasis in a design set(N=195)and a test set(N=66).A gene set enrichment analysis(GSEA)suggested that the risk score positively correlated with several cancer metastases-related pathways.Results: According to AFFY U133,the PLUS2.0 gene annotation file filters the probe to remove the probes that are annotated to multiple genes at the same time.The probe signal value with the highest average signal value was screened as the gene expression value.Then,the filtered average genes with exp ≥6 and max exp ≥7 were subjected to a subsequent analysis and were finally screened in the metastatic and nonmetastatic groups using a t test,with a p value≤0.05 for the genes(426 genes)used to straighten the CLM-related marker screening.Lasso’s regression algorithm was used to rank the importance of 426 genes.Genes CD163L1,FAM210 B,LGR5,LRRC16 A,PIK3R3,PLEKHA6,PROSER2,RBBP9,SEMA6 D,STOM,THBS1,and ZNF544 were potential markers.Real-time quantitative PCR showed the expression of FAM210 B and PIK3R3 in liver metastasis tissue was significantly higher than that in CRC tissue,and the expression of FAM210 B and PIK3R3 gene in liver metastasis tissue was significantly higher than that in normal intestinal tissue(P < 0.05).Further statistical analysis indicated that deregulation of FAM210 B and PIK3R3 suggest a poor prognosis for colorectal cancer liver metastasis.Conclusions: Biomarkers related to liver metastasis of CRC were screened by bioinformatics analysis of gene chip data,which provided basis for further study 。 12 genes including CD163L1,FAM210 B,LGR5,LRRC16 A,PIK3R3,PLEKHA6,PROSER2,RBBP9,SEMA6 D,STOM,THBS1,ZNF544 were screened as potential markers related to CRC metastasis.Protein-coding genes FAM210 B and PIK3R3 could be biomarkers or therapeutic targets for liver metastasis in colorectal cancer.
