Objective:1.In this experiment,a rat model of hyperlipidemia SD was established and a model of periodontitis SD rat was successfully established based on its model.2.The expression levels of TGF-β1,VEGF protein and mRNA in periodontal tissues of SD rats in each group were detected,and the correlation between periodontitis and hyperlipidemia was explored,which provided a meaningful reference for clinical.Method:1.Animal grouping and modeling: After adaptive feeding for 40 SD male rats for 1 week,they were randomly divided into four groups(n=10): group A: normal control group;group B: hyperlipidemia group;group C: periodontitis group;D Group: Hyperlipidemia + periodontitis group.Rats in groups A and C were fed with normal diet.Rats in groups B and D were fed with high-fat diet to establish a model of hyperlipidemia SD in groups B and D.The rats in groups C and D used 0.20 mm.A rat model of periodontitis was established by ligation of the left maxillary first molar combined with the smear method.2.Laboratory test:After 6 weeks of feeding on high-fat diet,four groups of rats were collected for serum lipids to determine the high-fat model.After 8 weeks of periodontal ligation,SD rat jaw samples were taken for Micro-CT scan to observe rat alveolar bone.Absorption;after 14 weeks of sacrifice,the periodontal tissues were decalcified by EDTA and dehydrated and embedded to produce histopathological sections.The periodontal tissue inflammation was observed to determine the establishment of periodontitis model.3.After the establishment of hyperlipidemia and periodontitis SD rat model,serum TGF-β1 and VEGF protein expression levels were detected by ELISA.The expression of TGF-β1 and VEGF mRNA in rat periodontal were detected by RT-PCR.4.Statistical analysis: SPSS20.0 statistical software was used.The data between the two groups were compared by t test or rank sum test.The comparison between multiple sets of data was analyzed by ANOVA or rank sum test.The test level was α=0.05,continuous variable.If it is normal,it is expressed by mean ± standard deviation,and the two pairs of comparisons between groups are analyzed by SNK method.Result:1.Hyperlipidemia model: The levels of serum TC,TG and LDL-C in group B were higher than those in group A at 6 weeks after high-fat diet.The level of HDL-C was lower than that in group A(P<0.05).There were no statistically significant differences in blood lipids in group A(P>0.05).There were no significant differences in serum lipids between group D and group B(P>0.05).2.Modeling of periodontitis: After periodontal ligation of 8W,the periodontal status of the four groups of rats was observed.There was no significant change in the gingiva of group A and group B.The periodontal probing was normal.The HE staining showed that the epithelial adhesion was tight and the periodontal membrane collagen fibers were arranged neatly.In group D,the gingival epithelial nails were long,and a large number of inflammatory cells infiltrated in the lamina propria.The periodontal membrane fibers were disorderly arranged,and the height of the alveolar crest was reduced.Osteoclasts and bone lacunae were seen.The degree of alveolar bone resorption in rats of group A,B,C and D was detected by Micro-CT.The statistical result was D>C>B>A.The value of alveolar bone loss in group C was significantly higher than that in group A(P<0.05).There was a significant increase in alveolar bone loss between group D and group C(P<0.05).3.The levels of serum TGF-β1 and VEGF protein in the four groups were detected by ELISA.The levels of TGF-β1 in group C were significantly lower than those in group A.There was significant difference between group D and group BC(P<0.05).The VEGF protein level in BC group was higher than that in group A,and the D group was significantly higher than that in BC group(P<0.05).4.The expressions of TGF-β1 and VEGF in periodontal tissues of four groups were detected by RT-PCR.The expressions of TGF-β1 and VEGF mRNA in BC group were significantly higher than those in group A(P<0.05).The expression of TGF-β1 and VEGF gene in group D was higher than that in BC group(P<0.01).Conclusion:1.In this experiment,a 8-week periodontitis SD rat model was successfully established by ligating silk combined with sputum smear method.Rats were fed with specific high-fat diet,and a hyperlipidemia SD rat model was successfully established for 6 weeks.2.Micro-CT results suggest that higher blood lipid levels have significant alveolar bone resorption,increased trabecular bone area,and higher blood lipids may aggravate the progression of periodontitis.3.The expression of cytokines in hyperlipidemia with periodontitis was higher than that in patients with periodontitis and hyperlipidemia alone.Hyperlipidemia may promote the development of periodontitis to some extent.
