Study On The Related Indicators Of Pioglitazone On Human Bladder Cancer Cells And Normal Bladder Transitional Cells | | Posted on:2020-12-07 | Degree:Master | Type:Thesis | | Country:China | Candidate:Y Bai | Full Text:PDF | | GTID:2404330590965019 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | Objective: To investigate the effects of pioglitazone,a PPAR-gamma agonist,on the proliferation,apoptosis,invasion and migration of normal bladder transitional cell SV-HUC-1 and bladder cancer cell 5637,and the expressions of PPAR-gamma,beta-catenin and AP-1.Materials and Methods: The cultured normal bladder transitional cells SV-HUC-1 and bladder cancer cells 5637 were treated with pioglitazone for 24 hours,48 hours and 72 hours respectively.The morphological and growth changes of the cells were observed by electron microscopy.MTT and flow cytometry were used to detect cell proliferation inhibition and apoptosis.Realtime quantitative polymerase chain reaction(PCR)was used to detect the expression of PPAR-gamma in two kinds of cells.Western blot was used to detect the expression of beta-catenin and AP-1 protein.Transwell assay was used to detect the effects of pioglitazone on the migration and invasiveness of bladder cancer cell line 5637.The effects of PPAR-gamma agonist pioglitazone on the proliferation,migration and expression of related factors of normal bladder transitional cells and bladder cancer cells were investigated.Results:(1)The proliferation of normal bladder transitional cells SVHUC-1 treated with PPAR-gamma agonist pioglitazone was significantly inhibited compared with the control group at 24 h,48h and 72 h,and the apoptotic rate of normal bladder transitional cells SV-HUC-1 increased with the increase of pioglitazone concentration after 48 h,the difference was statistically significant(P < 0.05).There was no significant change in bladder cancer cells 5637.(2)The expression of PPAR-gamma in normal bladder cells SV-HUC-1 and bladder cancer cells 5637 increased with the increase of pioglitazone concentration,and PPAR-gamma was less expressed in bladder cancer cells 5637(P < 0.05).(3)The expression of AP-1 and beta-catenin in normal bladder transitional cells SV-HUC-1 was not significantly changed by Western blot.The expression of beta-catenin and AP-1 in bladder cancer cells 5637 was higher than that in normal bladder cells,and decreased with the increase of pioglitazone concentration(P< 0.05).(4)With the increase of pioglitazone concentration,there was no significant change in the number of Transwell ventricular foramen penetration in bladder cancer cells.CONCLUSION: 1.PPAR-gamma agonist pioglitazone can promote apoptosis of normal bladder cells and inhibit their proliferation,but has no effect on proliferation and apoptosis of bladder cancer cells.2.PPAR-gamma is less expressed in bladder cancer cells than in normal bladder cells.3.AP-1 and beta-catenin are more expressed in bladder cancer cells than in normal bladder cells.4.PPAR-gamma agonist pioglitazone can inhibit AP-1 and betacatenin.PPAR-gamma agonist pioglitazone did not affect the invasion and migration of bladder cancer cells. | | Keywords/Search Tags: | Peroxisome proliferator activated receptor gamma, Activat or protein 1, beta-catenin, Bladder cancer, Pioglitazone | PDF Full Text Request | Related items |
