Objective:To investigate the expression of microRNA-21 gene in retinal and sclera of form deprivation myopia C57BL/6J mice,and the effect of Bujing Yishi Tablet on it,provide theoretical basis for early Chinese medicine intervention treatment of myopia,and provide new for the prevention and treatment of myopia research direction.Methods:In this experiment,3 weeks old healthy C57BL/6J mice were used to prepare the form deprivation myopia model,and the right eye was used as the experimental eye for comparison.The random unevenness was divided into 3 groups:normal control group(no treatment for both eyes)),the model group(the right eye shape deprivation started at the 22nd day),and the Chinese medicine intervention group(the 22nd day of age began to form the right eye shape deprivation and the Chinese medicine was administered).The first stage of the experiment:the normal control group and the model group were intragastrically administrated with the same amount of distilled water once a day;the Chinese medicine intervention group was intragastrically administered with a lean vision suspension,the concentration was 54.6 mg/ml,and the gastric perfusion volume was 0.1 ml/10 g.The mice were randomly sacrificed in the Chinese medicine intervention group from 3 weeks to 43 days old.The mice in the whole group,the normal control group and the model group were each labeled as normal control group 1,model group 1 and Chinese medicine intervention group.The second phase of the experiment:the remaining control mice were labeled as normal control 2 groups(both eyes did not do any treatment),the model group mice continued to form deprivation in the right eye,and were randomly divided into 5 groups:model 2 group,low Chinese medicine,medium and high dose groups,positive drug control group;normal control group 2 and model 2 group continue to use the same amount of distilled water for gavage,once a day;Chinese medicine low,medium and high dose group to be supplemented with Jingjingyijing suspension Stomach,once daily,the concentrations were:27.3mg/ml,54.6mg/ml,109.2mg/ml,gavage volume 0.1ml/10g;positive drug control group was given mecobalamin tablet suspension,daily Once,the volume was 0.01 mg/ml,and the concentration was 0.1 ml/10 g.All the mice in this part were sacrificed for 3 weeks to 64 days of age.The diopter of the mice was measured before and after the two-stage experiment.After the mice were sacrificed,the axial length of each group of mice was measured.The microRNA-21 gene of the retina and sclera of each group was detected by qPCR detection.The amount of expression.Results:1.Statistical results of diopter:Before the experiment,there was no significant difference in diopter between the three groups(P>0.05).After the first phase of the experiment,the model 1 group induced myopia compared with the normal control group 1(P<0.05).The experimental group 1 group had higher myopia degree than the Chinese medicine intervention group.Statistical significance(P<0.05);after the second stage of the experiment,the model 2 group,the low,medium and high dose groups of Chinese medicine,the positive drug control group were deeper than the normal control group 2,the difference was statistically significant(P<0.05).The diopter of the model 2 group was deeper than that of the high-,medium-,and low-dose groups of the Chinese medicine and the positive drug control group,and the difference was statistically significant(P<0.05).2.Axial statistical results:After the first phase of the experiment,the axial length of the model group 1 was 0.31 mm longer than that of the normal control group(P<0.05).The axial length of the model group 1 was relatively higher than that of the traditional Chinese medicine intervention group(P<0.05).After the second stage of experiment,the length of the axial length of the model 2 group was higher than that of the normal control group 2(P<0.05);the length of the axial length of the model group 2 was relatively higher than that of the other groups(P<0.05);the positive drug control group and the Chinese medicine each dose There was no significant difference between groups and within the group(P>0.05).3.Retinal and scleral qPCR results:Retinal gene results:After the first phase of the experiment:the gene expression of the model 1 group was relatively lower than that of the normal control group 1,and the model 1 group was relatively reduced compared with the Chinese medicine intervention group,and the difference in gene expression between the groups was Statistical difference(P<0.05).After the second phase of the experiment:the gene volume of the model 2 group was lower than that of the high,medium and low dose groups of the traditional Chinese medicine,and the difference was not statistically significant.There was no significant difference between the groups of the traditional Chinese medicine,and the positive drug control group was higher than the other groups.The difference was not statistically significant.There was no significant difference between the normal control group 2 and the other groups.Scleral gene results:After the first stage of the experiment:the gene volume of the model 1 group was higher than that of the normal control group 1,and the gene volume of the Chinese medicine intervention group was lower than that of the model 1 group,and the difference between the groups was not statistically significant(P>0.05);After:the gene expression of model 2 group was higher than that of high,medium and low dose groups of Chinese medicine,the difference was not statistically significant.The expression level of low dose group of Chinese medicine was lower than that of high dose group of traditional Chinese medicine,and the expression level of positive drug control group was lower than that of traditional Chinese medicine.In the dose group,the expression level of the positive drug control group was lower than that of the model 2 group,and the normal control group 2 was lower than the model 2 group,and the difference was statistically significant(P<0.05).Conclusion:1.The method of form deprivation can induce axial myopia in C57BL/6J mice.In the process of form deprivation,Bujingyishi tablets can delay the development of myopia.2,retinal and scleral microRNA-21 showed differential expression related to myopia,with the development of myopia,the gene is up-regulated in the retina and sclera,which may be involved in the growth regulation of the eye’s retina and sclera.3,Bujing Yishi Tablet has a certain regulatory effect on the retina and scleral microRNA-21 of myopic C57BL/6J mice,which may have a certain regulatory effect on remodeling of retina and sclera.
