Objective:To investigate the anti-inflammatory effect of yinjia tablet(YJ-T)on chronic pelvic inflammation in rats.Materials and methods:SD rats were selected and divided into two parts.The rat model of chronic pelvic inflammatory disease was prepared by mechanical injury and mixed bacterial infection.Ten rats were selected and divided into two groups,the model group and the normal group,respectively.Blood and uterine tissues of the rats were taken 14 days after the modeling,and the rat model was verified by visual observation and pathological sections.Then 50 rats were randomly divided into 5 groups,namely,high-dose silver-coated nail tablets,high-dose silver-coated nail tablets,low-dose silver-coated nail tablets,aspirin group and model group.Then,drug intervention was given for 2 weeks by gavage,and blood and uterine tissue of rats were taken.The expressions of cytokines il-2,il-6,il-8 and il-10 in serum were detected by ELISA.The morphological changes of uterine tissue were observed by visual inspection.The pathological changes of uterine tissue were observed under light microscope.The expression of TNF-a in utero was detected by immunohistochemistry.Finally,statistical software SPSS22.0 was used,and the results were expressed as mean standard deviation(x±s).If it does not conform to the normal distribution,the H test,namely the rank sum test,is selected.P<0.05 was statistically significantResults:Part I:1.Macroscopic comparison between the model group and the normal group indicated obvious adhesion between the uterus and surrounding tissues in the model group.2.Pathological section observation showed that the morphology of uterine cavity in the model group was changed,the wall structure was damaged and incomplete,the mucosa was congested,and most of the mucosal epithelial cells were columnar with obvious hyperplasia.Some interstitial structures were destroyed and disordered,and numerous inflammatory cells and neutrophils were seen in lamina propria and muscularis.Intrauterine adhesions are visible.Part ⅱ:1.According to visual inspection,the uterus of the model group was slightly white in color and wrapped by surrounding tissues,with severe adhesion.In some rats,there were different amounts of liquid in the pelvic cavity.In the high dose group,obvious uterine hyperemia,thickening of uterine cavity,and slight adhesion with surrounding tissues were observed.The macroscopic view of rat uterus in the low dose group was similar to that in the model group,with obvious adhesion to surrounding tissues and a small amount of peritoneal effusion.The gross morphology of yinjia tablets group and aspirin group were similar to each other,the degree of uterine tissue hyperemia was relatively light,the adhesion with surrounding tissues was less,and the activity was acceptable.No inflammatory mass was observed,and no unknown liquid was observed in uterine cavity and abdominal cavity.2.Pathological section observation showed that the cavity wall of the high-dose silver-nail film group was orderly with normal morphological structure,which was basically close to that of the normal group.The morphology and distribution of cells in the mucosa were basically normal,with some slight hyperemia and no cell degeneration and necrosis observed.In the yinjia tablet middle dose group and the positive drug aspirin group,some epithelial cells protruded into the uterine cavity,showing columnar hyperplasia,some mucosal epithelial structures were incomplete,inflammatory cell infiltration could be seen in the muscular layer,and hyperemia and enlargement could be seen in some intima.In the low-dose group,the structure of uterine cavity wall was incomplete,and the degenerative and necrotic parts were more obvious than those in the other groups.The epithelial cells were increased and showed a high columnar shape,and the intima showed hyperemia and edema.Inflammatory cells and neutrophil infiltration could be seen,and some of them even involved the muscle layer.See incomplete wall structure model control group,the mucosa lamina propria structure change,thinning or disappear,visible epithelial hyperplasia of dense,uterine cavity bump,the columnar to mucosa epithelial cells visible degeneration,necrosis,fall off,part of interstitial structure was damaged and contorts the disorder,muscular layer have a large number of neutral cells and inflammatory cells infiltration;3.There was no significant difference in serum il-2,il-6,and il-8 before administration.After drug treatment,the expression of serum il-2 in each dose group and aspirin group was lower than that in the model control group,and the difference was statistically significant(P<0.05).11-6 was significantly decreased after treatment in the traditional Chinese medicine dose group and aspirin group compared with the model group,with statistically significant differences(P<0.05).Il-8 was significantly decreased after treatment in the traditional Chinese medicine group and aspirin group compared with the model group,and the difference was statistically significant(P<0.05).After treatment,il-10 was significantly increased in each dose group of traditional Chinese medicine and aspirin compared with the model group,and the difference was statistically significant(P<0.05).After the treatment,the serum levels of il-2,il-6 and il-8 in the traditional Chinese medicine group were compared with those in the aspirin group,and the results showed that the expression of il-10 in the traditional Chinese medicine group and il-6 in the low-dose Yinjia group were statistically significant(P<0.05),while the rest were not statistically significant(P>0.05).4.Measured by means of immunohistochemical tumor necrosis factor alpha(TNF alpha)expression in rat uterus tissue,the results suggest the drug after intervention,in x200 and x400 then prompt the TNF alpha in every dose group of traditional Chinese medicine(TCM)model and the expression of positive drug aspirin group were lower than control group,each dose group compared with aspirin is found that high doses of silver nail group in 200 and 400 times of expression were lower than that of aspirin group.Conclusion:1.The rat model of chronic pelvic inflammatory disease was successfully established by mechanical injury plus mixed bacterial fluid infection 2.The anti-inflammatory effect of YJ-T may be achieved by reducing the expression of proinflammatory cytokines il-2,il-6,il-8 and TNF-α in the serum,while increasing the expression of anti-inflammatory cytokines il-10 in the serum.3.The ability of YJ-T to regulate inflammatory factors is closely related to its concentration.Overall,the YJ-T medium dose group had better effects in all aspects. |
